Objective: Malignant lymphoma is one of the most common blood diseases that severely affects people's health. It presents many kinds of clinical manifestations. For complicated pathogenies, people put more and more emphasis on it. Along with the rapid developments in molecular biology, people have realized that genetic alteration is a probable cause of malignant lymphoma. The polymorphisms of some compounds existing widely in nature such as quinone and substances in human bodies play essential roles in DNA synthesis and methylation and their metabolic pathways might thus affect disease susceptibility. Many researchers have done relative work, but there is no definite interpretations. The aim of our study is to investigate the assiciations between the polymorphisms and the susceptibilities to malignant lymphoma, to analyze the pathogeny of malignant lymphoma from the angle of molecular biology , to provide theoretical evidence for effective preventions and treatments to maliganant lymphoma . Methods: This case-control study recruited 116 patients with malignant lymphoma and 191 unrelated healthy controls. All of the cases were pathologically confirmed. All these patients were all hospitalized in the Fourth Affiliated hospital of Hebei Medical University between 2001,9~2003,3.The healthy controls were unrelated individuals for physical examinations during the same period as the patients.All of the patients and controls were from Shijiazhuang city or its surrounding regions. The information of sex, age was obtained from patients and controls by interview directly after bleeding. The diagnostic criterion was based on the fifth edition of Internal Medicine and the seventh edition of Textbook of Paediatrics. The different stages of malignant lymphoma was based on An Arbor stage system. 5 ml of venous blood from each subject was drawn in Vacutainer containing EDTA and stored at 4℃. The genomic DNA was extracted within one week after bleeding using proteinase K digestion followed by a salting out procedure .We analyzed the allele and genotype frequencies using PCR-RFLP. In the meantime, the cases were divided into several groups according to different characters such as clinical stages and pathological types. Genotype frequencies and allelle frequencies were compared in the patients and the controls using Chi-squared (X2) analysis on 2×2 and 2×3 cintingency tables and logistic regression. A P value of less than 0.05? was considered to be significant. Results: 1 There was no siganificant difference in sex,age and mean age between patients and healthy controls.The patients and the controls were comparable(P>0.05). 2 NQO1: The genotype distribution in patients and controls was not relative to age and sex. There was no statistic difference in allele distribution between patients and controls (P>0.05). The overall distribution difference was not observed between cases and controls(P>0.05). The genotype alone or combined genotypes also didn't show significant influence on the risk of developing malignant lymphoma(P>0.05). When stratified for clinical stages and pathlogical types, the significant difference was not seen either between patients in early stage and patients in terminal stage or between T and B cell malignant lymphoma patients(P>0.05). 3 TS : The genotype distribution in patients and controls was not relative to age and sex(P>0.05). There wasn't significant difference in allele or genotype distribution between patients and controls(P>0.05). The genotype alone or combined genotypes also didn't show significant influence on the risk of developing malignant lymphoma(P>0.05). When stratified for clinical stages and pathlogical types, the significant difference was not seen either between early and anvanced stage or between T and B cell malignant lymphoma patients(P>0.05). 4 MTHFR: The genotype distribution in patients and controls was not relative to age and sex(P>0.05).
