Administration of agent in pocket is one important therapy for chronic periondotitis. Carboxymethyl Chitosan, possessing a fairy excellent biocompatibility, is an non-toxic and non-stimulating material. Ornidazole is a new artificialanti-anaerobic agent with a high efficiency in inhibiting oral anaerobic bacterial. To study the effect of the topical application of a dental gel of the combination Ornidazole and Carboxymethyl Chitosan in the treatment of chronic periodontitis is of clinically significance.ObjectivesTo evaluate the biocompatibility and the bacteriastasis efficiency of Carboxymethyl Chitosan(CAMS) and to explore the feasibility of adoption of a Ornidazole of Carboxymethyl Chitosan dental gel in treatment of periodontical probing in clinic. Materials and Methods:1 Minimum inhibitory concentration (MIC) and biological compatibility.1.1 Twelve international standard oral anaerobic species, Porphyromonas gingivalis ATCC33277, ActinobacillusActinomycetemocomitans Y4, Prevotella intermedicus ATCC25261, Capncytophaga gingivalis ATCC 33624, Capncytophaga ochracea ATCC 27872, Prevotella melaninogenicus ATCC25845, Candidaalbicans BH829, Porphyromonas endodontalis ATCC 3 5 406, Prevotella densicola ATCC33185, Streptococcus mutans C, Streptococcus sobrinus ATCC27607, Actinomyces viscosus ATCC 19246, were selected as the target. Minimum inhibitory concentration (MIC) of Carboxymethyl Chitosan, Ornidazole and Carboxymethyl Chitosan Ornidazole was measured by gradient dilution method.1.2 0.2ml Carboxymethyl Chitosan, Ornidazole of Carboxymethyl Chitosan and dissolvable amylum were injected into the thigh of mouse respectively. The mouse were sacrificed at 1, 4, 7, 14 and 21 days after injection. The muscles of injected region, heart, spleen, liver and kidney was obsevered with microscope and transmission electrical microscope .1 A randomized, single blind, controlled clinical trail was performed. One hundred probing site, which had at least 5mm of Probing Depth (PD) on the symmetric quadrant were enrolled in this random study with split-mouth design. In the first group, a dental gel was administrated topically after subgingival scalling, while in the second group no agents was adopted after subgingival scalling. Plaque index (PLI) , bleeding index(BI), probing < Depth(PD) and subgingivalplaque microscopy were recorded before and 25 days after the ending of the treament. Results1.1 The MICs of Carboxymethyl Chitosan on Porphyromonas gingivalis ATCC33277, Actinobacillus Actinomycetemocomitans Y4, Prevotella intermedicus ATCC25261, Capncytophaga gingivalis ATCC 33624, Capncytophaga ochracea ATCC 27872, Prevotella melaninogenicus ATCC25845, Candidaalb leans BH829, Porphyromonas endodontalis ATCC3 5406, Prevotella densicola ATCC33185, Streptococcus mutans C, Streptococcus sobrinus ATCC27607, Actinomyces viscosus ATCC 19246, was respectively 20, 10, 5, 80, 20, >80, 20, 5, 20, 10, 60, 40mg/ml. The MICs Ornidazole on Porphyromonas gingivalis ATCC33277, Actinobacillus Actinomycetemocomitans Y4, Prevotella intermedicus ATCC25261 and Porphyromonas endodontalis ATCC35406 was respectively 5% 40% 20^ >160(ig/ml.The MICs Ornidazole of Carboxymethyl Chitosan on Porphyromonas gingivalis ATCC33277, Actinobacillus Actinomycetemocomitans Y4, Prevotella intermedicus ATCC25261 and Porphyromonas endodontalis ATCC35406 was respectively 1.25 > 20> 80> 40(ig/ml.1.2 Inflammation of the mouse in Carboxymethyl Chitosan test was mild. In contrast, there were a great deal of lymphsises infiltrate, multinucleated gant cell and evenobvious suppuration in amylum group. The inflammation in Ornidazole Carboxymethyl Chitosan group was at middle.
