Objective:The purposes of this project include: to explore the activated basophils expression ratio of acute, chronic urticaria patients and healthy controls peripheral blood. Contrast the expression ratio before and after allergen stimulation, to study effect of allergen on CD63 expression ratio and across hypersensitivity of urticaria patients, and to offer immunology method to detect allergen.Methods:The patients with acute and chronic urticaria were screened for those sensitive to allergens by scratch test with Der P, Der F and Cat E allergens. A comparative study on sensitivity to allergens was performed between the patients with acute, chronic urticaria and healthy individuals. Expression of CD123, CD63, HAL-DR was detected by three-color immunofluorescence flow cytometry. After allergen stimulation, the change of CD63 expression was analysed. Effect of allergen on basophils and crossed hypersusceptibility was studied.Results:The percentage of CD63+CD123+HLA-DR" basophils of healthy controls was significantly lower than those of the acute urticaria patients group and chronic patients. Moreover, the percentage of activated basophils in acute patients is higher than that in chronic ones.With Der P stimulation, the change of CD63 percentage of healthy controls wasnot obvious; however, urticaria patients' CD63 expression was up-regulated significantly. The other allergens had similar effect. Every patient has various activation level and ability.Six(50%) chronic urticaria patients and eight(66.7%) acute urticaria patients were hypersensitive to more than one type of allergen.Conclusion:The present study shows that urticaria patients have abnormal basophils activation in peripheral blood. The percentage of CD63 is higher in urticaria than that of healthy controls. After allergen stimulation, CD63 expression ratio in the sensitive patients was up-regulated. CD63 expression may can used for the elevation of allergens in urticaria. Detecting allergen by FACS may redounded to urticaria patients treatment as a new way.
