AIM: To study expressions of extracellular matrix protein-1(ECM1)and hyaluronan(HA)in laryngeal carcinoma and their relationships with the growth, metastasis and angiogenesis of tumor. METHODS: Using antibody of ECM1 and HA-combining protein to discover their expressions in laryngeal tumors (total 33 cases). Using RT-PCR technology to test the existence of ECM1 mRNA in both fresh laryngeal tissues and its surrounding tissues (total 5 cases). RESULTS: ECM1 and HA are expressed in both benign and malignant laryngeal tissues, chiefly in the cellular plasm. But the expression of both ECM1 (P=0.002) and HA (P=0.032) is significant different between benign and malignant laryngeal tumors. Their expressions are stronger in malignant tissues than in benign ones. In malignant laryngeal tissue, the expression of ECM1 is associated significantly with MVD(24.50±5.96) (Spearman's rho is 0.505, P=0.019). However, the expression of HA is associated significantly with clinical stage of laryngeal carcinoma (Spearman's rho is 0.521, P=0.015), and more it is stronger in tissue of cases with metastasis or relapse than that without. The difference is significant(P=0.030). ECM1 mRNA is found in both laryngeal carcinoma tissue and its surrounding l tissue and it seemed no difference. CONCLUSIONS: ECM1 made an important role in the growth, metastasis and angiogenesis of laryngeal carcinoma. The elevation of total level of HA is associated with laryngeal carcinoma. The existence of HA made growth and metastasis of laryngeal carcinoma easy. The laryngeal carcinoma cell with high secretion of HA has higher potential of metastasis and privilege of survive. The more study of ECM1 and HA will benefit the treatment of laryngeal carcinoma and the judgement of its prognosis.
