Objective: To study the effects of leukemia inhibitory factor (LIF) on nerve regeneration on rat's sciatic nerve defect that was bridged by silicon tube. Methods: Sixty SD rats (weight 250-300g) were divided into two groups with the same amounts at random. 10mm nerve defects were made and bridged with silicon tubes on left sciatic nerve of all rats. LIF was injected into the silicon tubes in experimental group for a dosage of 50ul(0.5ug) while the same volume of Saline in the control group. The tissues of the regenerative sciatic nerve in silicon tubes were observed of each ten cases in one, two and three months after surgery in both group. Result: The silicon tube that filled with LIF successfully induced the sciatic nerve regeneration across a 10mm gap, one , 2, 3 months after surgery ,the electrophologicaland morphological observations showed better regeneration of the myelinated axons on the test groups. Meanwhile SFI,the muscle mass and cross section area of muscle fibers of test groups were better than those of control groups .Conclusion : LIF can improve the injured peripheral nerve axon regeneration .
