Experimental Study About Interveninent Effects Of The Decoction For Promoting Blood Circulation And Removing Blood Stasis On Myocardial Cellar Apoptosis In Dogs Of Acute Ischemic Myocardium

Objective.To investigate interveninent effects of five decoction for promoting blood circulation and removing blood stasis on Myocardial Cellar Apoptosis in Dogs of Acute Ischemic MyocardiumMethods: Thirty-five dogs were randomly divided into 7 groups: control group myocardial ischemic group ShiXiaoSan group XueFuZhuYu group> HuoLuoXiaoLingDan group TaoHongSiWu group and DanShenYin group.Besides the control group and myocardial ischemic group,each of the dogs was given decoction for 5 days.Then the left anterior descending coronary artery was ligated to make a model of acute ischemic myocardium.Finally, Changes in myocardial pathology, the SOD activity , the level of CPK ,LDH and MDA were detected. By TDT/d UTP Nick End Labeling and immunohistochemical method,we can observe themyocardial cellar apoptosis and protein'expressions of apoptosis regulatory gene Bcl-2,Bax in acute ischcmjc myocardial dogs .Results: The SOD activity of XueFuZhuYu group, HuoLuoXiaoLingDan group, TaoHongSiWu group and DanShenYin group were significantly higher than those of myocardial ischemic group and ShiXiaoSan group respectively,while the level of CPK ,LDH and MDA were significantly lower than those of myocardial ischemic group and ShiXiaoSan group respectively. Compare with myocardial ischemic group and ShiXiaoSan group, the number of myocardial cellar apoptosis in XueFuZhuYu group, HuoLuoXiaoLingDan group, TaoHongSiWu group and DanShenYin group were significantly less respectively. The number of myocyte necrotic cells increased in myocardial ischemic group and ShiXiaoSan group respectively.Conclusion XueFuZhuYu decoction , HuoLuoXiaoLingDan decoction, TaoHongSiWu decoction and DanShenYin decoction can raise the SOD activity,lower the level of CPK ,LDH and IMDA,affect protein' expressions of apoptosis regulatory gene Bcl-2,Bax and prevent myocardium cells from apoptosis.Therefore,these four ones play a role of protecting ischemic myocardium.The mechanism may be related to their own characters of decoction. XueFuZhuYu decoction , HuoLuoXiaoLingDan decoction, TaoHongSiWu decoction and DanShenYin decoction put an emphasis onactivating qi as well as promoting blood circulation.However, ShiXiaoSan decoction underlines promoting blood circulation at expense of activating qi.This research shows there are several factors which lead XueFuZliuYu decoction, DanShenYin decoction, HuoLuoXiaoLingDan decoction and TaoHongSiWu decoction to preventing myocardial cells from apoptosis.The factors as follows: inhibiting enzyme from outflowing, eliminating oxygen free radical and affecting protcin'expressioius of apoptosis regulatory gene Bcl-2,Bax.
