Objective:To explore the therapeutic effect and mechanism of Fangfeng and Taohong Siwu decoction on hepatic fibrosis in rats,as well as the synergistic effect of Fangfeng on promoting blood circulation and removing blood stasis,to provide a new strategy for the treatment of hepatic fibrosis.Methods:fifty clean male SD rats were randomly divided into normal control group?group A?and model group.The rats in the model group were intraperitoneally injected with 40%carbon tetrachloride?Carbon tetrachloride,CCl4?soybean oil solution,and the rats in the normal control group were given the same amount of normal saline twice a week for 8 weeks.After 8 weeks,2 rats in the normal control group and 4 rats in the model group were injected intraperitoneally with chloral hydrate.The liver of the rats was extracted and stained with hematoxylin-eosin?Hematoxylin and eosin,HE?and Masson.The rats in the model group were randomly divided into pathological model group?group B?,colchicine group?group C?,Fangfeng Taohong Siwu decoction group?group D?,Taohong Siwu decoction group?group E?and Fangfengshui decoction group?group F?.The rats in group A and group B were given the same amount of normal saline,and the other groups were given the corresponding drugs.Except for group A,the rats in the other groups were injected with 40%CCl4 soybean oil solution intraperitoneally until the end of the experiment.Four weeks in a row.After the last administration,the blood samples of abdominal aorta were collected and the serum was taken.The liver,spleen and kidney tissues of rats were extracted and weighed.Detection of glutamic oxaloacetic transaminase?AST?,glutamic pyruvic transaminase?ALT?,alkaline phosphokinase?ALP?,hyaluronic acid?HA?,laminin?LN?,procollagen-3-peptide?PCIII?andCollagen in Serum?IV-C?.Detection of Transforming growth Factor-?1?TGF-?1?,alpha-smooth muscle actin??-SMA?and B Lymphoma-2 Gene?Bcl-2?by Western blot?WB?.The expression of Bcl-2-associated X protein?Bax?was detected.The expressions of TGF-?1 and?-SMA in liver tissues were analyzed by immunohistochemistry.Results:?1?after 8 weeks,the HE staining of liver tissue in the model group showed congestion of central vein,disordered arrangement of hepatocyte cord,many pathological areas of liver tissue,destruction of hepatic lobular structure,and degeneration and necrosis of hepatocytes in some liver tissues.Masson staining showed that the structure of hepatic lobule was destroyed,a large number of collagen fibers were deposited in some areas,and collagen fibers were deposited in the surrounding and portal areas to form fiber septum.?2?after 4 weeks of treatment,compared with group B,the indexes in serum and the expression of Bax,TGF-?1 and?-SMA protein in liver tissue were significantly decreased in group C,group D,group E and group F?P<0.05?.The expression of Bcl-2 protein was significantly increased?P<0.05?.Compared with group C,the expression of ALT,AST,HA,PCIII,IV-C in serum and the expression of Bax,TGF-?1 and?-SMA protein in liver tissue in group D were significantly decreased,while the protein expression of Bcl-2 was significantly increased?P<0.05?.There was no significant difference in the levels of ALP and LN,and there was no significant difference in the changes of detection indexes between group E and group F.Conclusion:?1?Fangfeng Taohong Siwu decoction can improve the indexes of ALT,AST,ALP,HA,PCIII,IV-C liver function and liver fibrosis in rats.Its effect is better than that Taohong Siwu decoction and Fangfeng decoction alone;?2?Fangfeng has the effect of promoting blood circulation and removing blood stasis;?3?Fangfeng Taohong Siwu decoction can improve liver fibrosis.The mechanism may be related to the regulation of Bcl-2/Bax apoptosis pathway and the down-regulation of TGF-?1and?-SMA protein expression. |
