It has been for a long time to use Mu-tong on the treatment of stranguria (as urinary tract infection in modern medicine) in TCM clinical practice. Since Guan-Mu-tong(Caulis Aristolochia manshuriensis) has been unused for its adverse incidents, Mu-tong(Caulis Akebiae) will be reused as main variety. We will study on analyzing the scientific possibility of substitute Caulis Akebiae trifoliate for the poisonous Caulis Aristolochia manshuriensis from both sides of safety and curative effect in the first step, in order to provide scientific experimental basis for clinical use, and method reference for the Chinese herbal pharmacology and toxicology.Firstly, imitate the onset of human clinical urine tract infection, (the point is that pathogenic organism principally E. coll. from urethra up to urinary bladder, then from bladder with urine adverse current to kidney, and the necessary environment of E. coll attachment), under the condition of acidification of bladder membrane, use the method that by pipe conversely inject the right amount of E.coli., establish non-surgical model of urine tract infection in domestic rabbit, comprehensive evaluate various index of bacterial culture in urine and kidney, measurement of frequency of micturition and volume of urine, urine tract histology and pathology changes, general physiology, biochemistry, biology characteristics such as the combine with body temperature, weight, food and water intake, blood and urine routine examination and survival rate etc, confirming the model is successful, provide an experimental method closer to clinic for observing the pharmacological function of Caulis Akebiae.Then, primarily observe diuresis and bacteriostasis of Caulis Akebiae trifoliate on the above model, and compare with Caulis Aristolochia manshuriensis. The results show that animals in group of Caulis Akebiae trifoliate are stable in general situation, with growthy status and physiological activities, without death by poisoning; taking Caulis Akebiae trifoliate for about a week by ig., bacteriuria has a decreased trend, without effects on frequency of micturition and volume of urine. However, animals in group of Caulis Aristolochia manshuriensis are abnormal in general situation, withloss of body weight (P0.05) and fall of body temperature, presenting glycosuria, damage and necrosis in renal tubule, more severe histologic and pathologic changes, and large number of animals death by poisoning; after taking Caulis Aristolochia manshuriensis by i.g., bacteriuria not only decreasing but also increasing, volume of urine first up then down. The results indicate that Caulis Akebiae trifoliate is safety without poisonous, and has a trend of bacteriostasis; but Caulis Aristolochia manshuriensis is proved to be poisonous, with increasing bacteriuria and aggravating histologic and pathologic process, accelerating death of a large number of animals. Therefore, it is safety and valid that substitute Caulis Akebiae trifoliate for Caulis Aristolochia manshuriensis.Finally, by slip method, observe bacteriostasis of Caulis Akebiae trifoliate in three doses of high, middle and low in vitro, while compare with Caulis Aristolochia manshuriensis. The results show that both Caulis Akebiae trifoliate and Caulis Aristolochia manshuriensis have bacteriostasis in different degree, and the bacteriostasis of them are stronger in low doses than in high and middle doses, however, the potential of bacteriostasis of Caulis Akebiae trifoliate is stronger in vitro, The results not only verified the same bacteriostasis of Caulis Akebiae trifoliate in vivo and in vitro, but also further indicate that the mechanism on bacteriostasis of Caulis Akebiae trifoliate may be concerned with directly repressing to E.coli.. Moreover, the different results of Caulis Aristolochia manshuriensis on bacteriostasis in vivo and in vitro suggest that it may be concerned with body failure and hypoimmunity caused by nephrotoxicity of Caulis Aristolochia manshuriensis.In brief, this study provide experimental basis of substitute Caulis Ake...
