BACKGROUNDS AND AIMS The pancreatic exocrine secretion is an important component in thepostpradial digestion and absorption of food. It is thought that this courseis mainly regulated by endocrine hormone cholecystokinin (CCK) andvago-vagal reflex at present,which act on vagal postganglionic cholinergicneurons of pancreas. But how the signals of vagus nerve uploaded andintegrated in the nerve center after the meal, and then unload to controlpancreatic secretion and what neurotransmitter participates this pathremainsl unclear. The nerve center's regulation of the pancreatic exocrinesecretion mainly depends on the parasympathetic nerve sent out from thedorsal motor nucleus(DMN) . It has already proved that there is anexciting or inhibitory effect on the pancreatic exocrine secretion functionwhen some neurotransmitter microinjected into the dorsal motor nucleus,but how the center spreads the signal to the pancreas to mediate thepancreatic exocrine secretion and what is the relevant neurotransmitterremains unclear. In this study we investigated the influence ofintraduodenal infusion of 20% intralipid on the activity of intrapancreaticneurons by expression of c-Fos in intrapancreatic neurons and observed theinhibitory effect of microinjection of somatostatin( SS ) into the dorsalmotor nucleus on the basal pancreatic exocrine secretion. We hope thisstudy can strengthen the understanding on the mechanism of how luminalstimulations mediate pancreatic exocrine secretionin this 6第二军医大学硕士学位论文 英 文 摘 要investigation,and strengthen the understanding on hypothesis ofBrain-Pancreas Interaction.And this study has an important influence onthe treatment and cure of the pancreatic disease.SECTION ONE SETTING-UP OF THE ANIMAL MODEL TO STUDY THE PANCREATIC EXOCRINE SECRETION FUNCTION IN SD RATSObjective: To establish the animal model to study the pancreatic exocrinesecretion function in healthy male SD rats. Methods: SD rats weighing250~300g were selected,after anesthetized the rat was equipped withpancreatic duct cannula inserted into the sphincter of Oddi to collect thecombined bile-pancreatic secretions.A segment of small intestine, includingthe whole duodenum was isolated between the first intestinal cannulapositioned 5mm from the pylorus and the second intestinal cannula at theboundary of duodenum and jejunum to permit the intraduodenal infusion.The pylorus was ligated. Results: After equipped with pancreatic ductcannula, there were clear yellowish combined bile-pancreatic secretionsflowing out from the pancreatic duct cannula. The volume of the combinedbile-pancreatic secretions after the duodenal infusion of 20% intralipid wasincreased by 54.17%. Conclusion: We succeed in setting up the animalmodel to study the pancreatic exocrine secretion function in SD rat.
