| OBJECTIVE: Endometriosis (EM) is an estrogen-dependent disease characterized by the presence of endometrial glands and stroma outside the uterine cavity. The etiology of this disease remains elusive, but is clearly influenced by endocrine factor. There is no the most effective and feasible therapy for endometriosis. The aims of our study were to investigate the changes of reproductive endocrine of EM and the effect of Neiyi recipe through clinical and animal experimentations.METHODS: In clinical experimentation, blood samples were collected from fifty women (between the ages of 27and 41) with EM and fifty normal reproductive-age women (between the ages of 18 and 35) during the menstrual cycle. The levels of peripheral blood FSH, LH, E2, PRL, PGE2 and the luteal phase P were detected by radioimmunoassay (RIA). In animal experimentation, Estradiol Benzoate (EB)-primed ovariectomized (OVX+EB) mouses were injected intravenously (i. v.) with PGE2 (Img/kg) or physiological saline. The mouses were killed in 15, 30, 60, and 90 minutes after injection. Hypothalanius was separated and weighted. Concentrations of GnRH and NPY were determined by RIA. Experimental EM rats were used to study concentrations and correlation of PGE2 and reproductive endocrine with or without treatment of Neiyi recipe.RESULTS: Peripheral blood plasma PRL concentration in EM patients was significantly elevated than that hi control group in the luteal phase (P<0.05). The level of plasma LH was lower in EM patients than that hi control group in the menstrual phase and follicular phase (P<0.05). There was no difference in the level of FSH, E2 and P between the EM patients and control group (P>0.05). Peripheral blood serum PGE2 concentration in EM patients was significantly higher than that in control group, except the luteal phase (P>0.05).Concentrations of hypothalamic GnRH and NPY in EM rats were lower than that in sham-operated control (P<0.05). Hypothalamic GnRH after injection of PGE2 released more significantly than saline, reaching a peak at 30 minutes. There was a postive correlation between PGE2 and PRL in the luteal phase (r=0.80, P<0.05) of EM patients. There was a postive correlational, P<0.05) between peritoneal fluid PGE2 and pituitary PRL in EM rats without treatment. Concentrations of peritoneal fluid PGE2, serum E2 and pituitary PRL of endometriosis rats were significantly lower after treatment of Neiyi recipe, increasing levels of hypothalamic GnRH and NPY (PO.05). Implanted endometrium was reduced after treatment of Neiyi recipe.CONCLUSIONS: EM existed in the dysfunction of reproductive endocrine. PGE2 might play an important role in the dysfunction of reproductive endocrine of endometriosis patients. Neiyi recipe could regulate the dysfunction of reproductive endocrine by hypothalamic-pituitary-ovarian axis, improved function of ovary, reduced ectopic endometrium. |
