Objective:To study the pathogenesis of ulcerative colitis(UC) and the main mechanism of KuiJieAn(KJA)in protection of UC in rat. Methods :A rat model of UC with the TCM syndrome of "Liver depression and SPleen deficiency" was induced by electric stress, taking food irregularity and acetic acid . In order to detect the effects of KJA, The macroscopical examination of the colon were evaluated. The function of antibody-mediated immunity, cell-mediated immunity, the lymphocytic subtype in peripheral blood and TNF— α , IFN-γ, IL-4 in serum were analyzed. Then detected the levels of gut hormone , the activity of SOD, GSH—PX and the concentration of MDA, NO. Result: It showed that KJA can increase the function of antibody-mediated immunity, decrease the hyperfunction of cell-mediated immunity, regulate the subtype of lymphocyte by suppresses of increased TNF—α , IFN-γ and raise the decreased IL-4. Both the level of gut hormone and MDA, NO were decreased significantly. The activity of SOD, GSH—PX were raised. Conclusion:The data suggest that the cell mediated inflammatory reSPonses and antibody-mediated immunity as important mechanism of forming the UC. It can be inferred that pathogenesis of UC is correlated closely with confusion of immunological function and moodfactor. Meanwhile, KJA has protective effects on UC of rat by maintaining the balance of Th1 cell and Th2 cell, regulate the system of neurological-endocrinological-immunology and have the effect of antioxygenic action .
