Objective: Evaluated the difference of life expectancy ,quality of life and tumor weight of neoplasitc cases with depression during different tumor-stage by animal experiments observation, to reveal the affection of depression on malignant tumors and it's possible immunity mechanisms. Methods: 1,Divided rats stochasticly into four groups named A,B,C,D, and developed the animal model as stimulations of depression associated with tumor by separation feeding and chronic unpredictable mild stress before/after their receiving tumor-cell-inoculation. Observed life expectancy ,quality of life and tumor weight of neoplasitc rats under different depression severity and suffering time. 2,The depressive behavior and status of rat was detected by open-field test controlled by computer,concentration of 5-HT in hippocampus measured by spectrophotofluorometric method ,and sweet-water-wastage test. 3,The impression that the blood serum of neoplasitc rats inhibited normal lymphocyte transformation introduced by ConA and augmentation of natural killer (NK) killing activity from neoplasitc rats studied in vitro was mensurated by MTT assay among different depression conditions. 4,Tumor necrosis factor-alpha (TNF-a) level in serum was measured by an murine L929 bioassay on cellular supernatantsin order to reveal depression's role playing in malignant tumor progress. Results: 1,evaluation of depression:①open-field test: There were no obvious changes in active time,rest time,center active time and total journey(p>0.05). After mild stress, active time, center active time and total journey reduced obviously while rest time obviously prolonged(p<0.01). ②The quantities of sweet-water-wastage for four groups were 34.0±7.8ml(A),20.5±8.6 ml(B) ,18.7±9.4 ml(C) ,9.3±3.42 ml(D). With depression developing, the sweet-water-wastage reduced(F=31.828,P<0.01).The quantity o"fD"group was the lowest while it of"A"group was the highest.there was no difference between that o"fB"group and"C"group(P=0.434).③The concentration of 5-HT in hippocampus declined when depression developed(F=45.816,P<0.001). The quantity o"fD"group was the lowest while it of"A"group was the highest.There was no difference between that of"B"group and"C"group(P=0.847). 2,KPS and tumor progress:At the first stage of"A"and"B"group after being inoculated withWalker-256,there were no obvious changes in spirit, activity, diet and body configuration. At 10th day postinjection, mice became thin, their activities were slower than before and spirit became worse.Drinking and eating reduced obviously.The mice status of"B"group were worser than that of"A"group. The spirit and activity of mice in"C"and"D"group were a little worser than that o"fA"group. At 10th day postinjection, there were much more obvious changes in spirit, activity, diet and body configuration of mice in"C"and"D"group. The mice status of"D"group were the worst. There was no significant difference found among four groups of tumor-emergence-time and tumor weight.3,The neoplasitc rats'average life expectancy of"D"group was obiously lower than that of"A"group(P<0.05). There was no significant difference found among four groups of life expectancy ( F=1.547 ,P=0.233).4,The blood serum of neoplasitc rats in four groups obviously inhibited normal lymphocyte transformation introduced by ConA.The affection"D"group was the most significant while that of "A "group was the weakest(P<0.01).The difference of inhibition role existed among four groups(F=11.961,P<0.001),but didn't exist between"B"group and"C"group(P=0.486).5,When tumor necrosis factor-alpha (TNF-a) level in serum was measured,we found that it of"A"group was obviously lower than that of the other three groups(P<0.05).We didn't find it significantly different among three groups(B,C,D) (P>0.05). The result appeared that TNF-a level in serum related with depression,but didn't relate with the time depression developing and wheather depression existing before or after cancer.6,The four groups significantly differed from NK cells killing activity( F=5.198,P=0.001). "A"group showed the hightest activity among the four groups, while"D"group acted the lowest activity".B"group was similarto"C"group in NK cells killing activity(P>0.05). Conclusions: 1,The experiment is the first to develop an animal model for studying the affection of depression on malignant tumors successfully. At the same time we comfirmed different depression severity and suffering time declined the quality of life and life expectancy of neoplasitc cases through animal experiment. 2,It realized in the animal experiment that chronic mild stress resulted in depression.The model rats'behaviors and characters such as interesting lost,happiness deficiency and activity reducing were similar to that of depression patients.The animal model was perfect. 3,The concentration of 5-HT in hippocampus declined with depression . The concentration of 5-HT within brain appeared the same change tendency as NK cells killing activity. NK cells killing activity perhaps regulated by 5-HT of nerve centre. 4,The stress depression didn't impact the tumor weight of neoplasitc rats,but made their quality of life worse,reduced their life expectancy. 5,The stress depression impacted both cellar immunity and body fluid immunity of neoplasitc rats.The body's cellar immunity was inhibited and TNF-αlevel in serum increased.As a result, it played bad role in neoplasitc rats prostrate state.6,The impactive mechanisms for relationship between depression and malignant tumor closely related to neuroendocrine-immune modulation( NIM) system.
