According to statistical data from WHO,cancers are the second dangerious diseasesafter angiocardiopathy and cerebrovasular diseases,which are greatly harmful to humanhealth.Chemotherapeutics is a sort of principal treatments for cancers.But cancers are rarelyradically cured by chemotherapeutics,because chemotherapeutic agents restricted by theirblood concentrations and toxicity focus on the tissues of tumor very difficultly.In addition,tumor cells vary easily to drug resistance and are not concentrated on one kind ofchemotherapeutic agents,meantime,chemotherapeutics cause easily toxic and side effect onthe whole body.So it is of important significance for human health to explore a sort ofeffective treatments for cancers,to improve drug sensitivity for cancer cells and to reducenegative effects of chemotherapeutics.For that reason,we employed high voltage prickelectrostatic fields(HVPEF) and HVPEF with chemotherapeutics to treat tumor cells invitro,and studied effects of HVPEF and HVPEF with chemotherapeutics on the growth,proliferation and mortality of HeLa cells.We did two experiments.One was that HeLa cellsin vitro were only treated by HVPEF for 5 days continuously.The other was that HeLa cellsin vitro were treated by both HVPEF and chemotherapeutic agents 5-Fu and DDP(the lastdrug concentration was 1 μg/μl)for 5 days continuously.The treatment time was 30 min/deach group.We used 5 different voltages of 0 kV,2 kV,6 kV,10 kV and 14 kV in theexperiments.MTT method was adopted to detect cell vitality;Trypanblue rejection methodwas used to protract cell growth curve;FCM was employed to detect cell mortality rate.Theexperiment results were analyzed by Student't test. We obtained experiment results as follows:1.HeLa cell growth was affected only afterHVPEF.(1)The results of MTT and FCM showed that the effects of HVPEF on HeLa cellproliferations and mortality rates presented "Electrostatic Field Intensity WindowEffect".The cell proliferation was increased and cell mortality rate was decreased after 2 kVHVPEF.But the cell proliferation was inhibited and cell mortality rate was promoted after 10kV HVPEF.The results showed that tumor cells could be killed effectively by appropriateintensities of HVPEF.(2)The effects of HVPEF on HeLa cell proliferations and mortalityrates presented "Biology Window Effect".We saw the different effects of HVPEF on HeLacells at their different proliferation phases.The results of cell growth curves showed that allvoltages inhibited cell proliferation and prolonged cell multiplication time.So we suggestedthat HVPEF could effectively kill HeLa cells at their appropriate proliferation phases. 2.HeLa cell growth was affected after HVPEF with chemotherapeutics.(1)The resultsof MTT showed that cell vitality was declined after HVPEF with DDP.Especially cellvitality after 10 kV HVPEF with DDP was significantly decreased compared with only DDPcontrol group.The results showed that HVPEF could increase DDP toxicity to HeLa cellsand inhibit cell proliferation.(2)The results of FCM showed that HeLa cell mortality rateswere all raised after different HVPEF with 5-Fu and DDP,respectively.Thus we suggestedthat tumor cells might be more effectively killed after optimum HVPEF withchemotherapeutic agents than after only chemotherapeutic agents,that is optimum HVPEFcould increase sensitivity of chemotherapeutic agents to tumor cells.
