It' s well known that a pathologic hallmark of systemic lupus erythematosus(SLE) is the recurrence of widespread and diverse vascular lesion.To investigate the existence of endothelial injuries of SLE ,we selected 31 SLE patients and 10 normal controls to analyse the plasma levels of von Willebrand factor (vWF), souble thrombomodulin(sTM) by ELISA.The correlation analysis(pearson' s correlation) was performed between vWF,sTM and SLEDAI, the quantity of 24 hour urine protein to investigate the relationship between endothelial cell injury and disease activity , renal lesion.To investigate the the prevalence of anti-endothelial cell antibodies(AECA) in SLE patients and normal controls,we detected AECA by indirect immunofluorescence assay and assessed the difference between the two groups.Two-color cytometry was applied to detect the expression of vascular cell adhesion molecule-1(VCAM-1) , E-selectin in the membrane of endothelial cell line ECV-304 cells,which were cultured with sera from SLE and normal control to study the role of AECA, VCAM-1, E-selectin in damage mechanism of endothelial cell in SLE.
