Objective To explore clinical scale expansion of human bone marrow mesenchymal stem cells (MSCs) in serum free media and their immunosuppressive property in mixed lymphocyte reactions in vitro.Methods Human bone marrow MSCs were obtained from 12 healthy donors or patients with hematologic malignancies and MSCs were expanded in vitro in Mesencult serum free media. Lymphocytes were prepared from two healthy volunteers. MSCs were cocultured with one-way mixed lymphocyte reactions (MLRs) and bidirectional MLRs, responder cells were labeled with carboxyfluorescein diacetate-succinimidyl ester (CFSE) in bidirectional MLRs. Cell Counting Kit-8 ( CCK-8 ) kit was used in cell proliferative detection, T-cell subsets were analyzed by flow cytometry (FCM).Results Human MSCs were isolated from a mean + SD of 26.3 ±8.8 ml of bone marrow aspirates from all subjects.
