Objective:The purpose of this study is to discuss the effect and molecularmechanism of Angiotensinâ…¡ (Angâ…¡) on erythroid progenitors advantageddifferentiation.Methods: Cord blood from normal full term uterine-incision delivery fetus wasused. Mononuclear cells was separated and purified by lymphoid separatingmedium , then added to suspension medium and cultured with Angâ…¡ anddifferent stimulant factors. After incubation, cells were transferred tomethylcellulose semisolid medium and cultured fourteen days. Theconformation of CFU-GM and burst-forming unit-erythroid- (BFU-E) colonieswere observed and the numbers of colonies were counted. At the same timeflowcytometry was used to detect the proportion of erythroid and myeloid.Nested RT-PCR was used to detect the expression of angiotensin II typelreceptor (AT1R) mRNA.Results: The expression of AT1R mRNA was detected at days 6 and 9 in theerythropoietin group; Ang II induced the differentiation and proliferation of cordblood progenitors to erythroid progenitors.Conclusion: Angiotensin II had stimulatory effect on erythroid progenitors invitro and this effect was mediated through its AT1 receptor.
