Objective: To investigate the feature of (99m)~Tc-HL91 hypoxic imaging on tumor and mechanism of action of anti-angiogenic agent thalidomide, moreover, to discuss the possibility of using (99m)~Tc-HL91 to monitor response to Thalidomide for Tumor of Mice. Methods: ⑴The prepared bearing-tumor mice model. Lewis lung cancer cell concentration was regulated to 5×107/ml. Lewis tumors were grown from 1×107 (0.2ml)cells inoculated subcutaneouly in right lateral hind limb of 40 C57BL/6 mice. H22 liver cancer cell concentration was regulated to 5×107/ml too. H22 tumors were grown from 1×107 (0.2ml) cells inoculated subcutaneouly similarly in right lateral upper limb of 10 C57BL/6 mice. ⑵Grouping and administration. The 40 mice inoculating lewis lung cancer were divided randomly into 8 groups which contain 5 mice. The No.1, 3, 5 and 7 groups were experiment groups. The No.2, 4, 6 and 8 groups were control groups. The mice inoculated H22 liver cancer were divided randomly into 2 groups which named No.9 and No.10 group. The No.9 group is experiment group and the No.10 group is control group. The No.1, 3, 5 and 9 groups were administrated thalidomide 200mg/kg/d by intragastric administration, and the No.7 group was administrated thalidomide 400mg/kg/d by intragastric administration. The No.2, 4, 6, 8 and 10 groups were administrated NS equal to the volume of thalidomide administrated to experiment groups in the same way. ⑶The mice bearing tumor imaging and tumor section imaging. (99m)~Tc-HL91 hypoxic imaging studies were performed on the No.1 and No.2 groups when administrated for 5 days. (99m)~Tc-HL91 hypoxic imaging studies were performed on the No.3 and No.4 groups when administrated for 9 days. (99m)~Tc-HL91 hypoxic imaging studies were performed on the No.5 and No.6 groups when administrated for 13 days. (99m)~Tc-HL91 hypoxic imaging studies were performed on the No.7 and No.8 groups when administrated for 9 days. The No.9 and No.10 groups were performed the same study respectively when administrated for 9 days 21 days and performed tumor section imaging following the 21 day's imaging. ROI were drawn around tumor and contra lateral limb on mice bearing tumor images and radioactivity radios of tumor to contra lateral limb (T/L) were calculated. ⑷Anatomy and pathologe analysis. After each group's imaging completed, executed mice, dissected tumors, weighted them immediately and put them into 10% formalin as soon as we can. Then these tumors were made to specimen. All tumor sections were studied by means of HE staining and CD34 immunohistochmical staining and microscopy. Microvessel density (MVD) and thickness of livetumor cells layer were measured. Results: ⑴It isn't markedly different to tumor weight between experiment groups and corresponding control groups (group 1and 2, group 3 and 4, group 5 and 6, group 7 and 8, group 9 and 10, P >0.05) .⑵Each other is significantly different to tumor weight among experiment group 1, 3 and 5 by single factor analysis of variance and SNK-q test (F=143.87,P<0.01). Each other is significantly different to tumor weight among control group 2, 4 and 6 by single factor analysis of variance and SNK-q test (F=132.14, P<0.01). The tumor weight always increased with the time of administration and growth. ⑶Each other isn't significantly different to tumor weight among group 3, 4, 7 and 8 which were dissected at No.9 day (F=132.14, P<0.01).⑷It isn't significantly different to T/L between group 1 and 2 (P>0.05). It is significantly different to T/L between group 3 and 4 (P<0.05). It is more different to T/L between group 3 and 4 than before (P<0.01). Both experiment series and control series show T/L gradually increased in whole experiment period. Among group 1, 3, and 5(F=20.56, P<0.01), it isn't significantly different between group 1and 3, group 5 is significantly different to others. Among group 2, 4 and 6(F=33.89, P<0.01), it isn't significantly different between group 2 and 4, group 6 is significantly different to others. ⑸It is significantly different to T/L between group 7...
