Objective: The incidence of ulcerative colitis in Europe and America is higher than other area , but the amount of patients of UC in China has been 3.08 times , which has a trend of increasing in last 10 years . Though the initiating event and pathogenesis of UC remains unclear currently , more and more evidence show that cytokines play an important role in initiating and developing of UC . Recent investigations have focused on transcription factors which regulate production and activation of cytokines , especially on nuclear factor-kappaB(NF-κB). The exact role of these factors in UC is still unknown , further studies may lead to identification of additional therapeutic targets which could improve management of the disease . The therapies of UC are various and the curative effects reported are different . The therapies abroad have focused on how to weaken or prevent the action of cytokines. It was report-ed that preventing the action of cytokines could not be proved effective through extensive multicenter prospective clinical exp-eriment . Though using inhibitors together might improve the curative effects , it is not feasible in practice obviously . NF-κB is a commonly existed transcription factor and the focus of tran-sduction of many signals , whose activation and metastasis into nucleolus is the premise and key point of cytokines'activation and deliverance . People have payed more attention to the study of NF-κB , it has been one of the most active research fields . The main domestic therapies were the application of anti-inflammatory medicines, traditional Chinese medicine and the combining traditional Chinese medicine and Western medicine , the effects of the last two therapies were reported better than th-at of the first one . The aim of this study was to investgate the effects of tradit-ional Chinese medicine kuijietang(KJT)in UC and influences on NF-κB by mensurating a series of objective parameters be-tween control groups , and to probe into the therapeutic mechan-ism of KJT . Methods: 1.Choice of study objects: Choosing 60 patients who conform to the standard . 2. Patients groups distribution and treatment: Dividing 60 patients into 3 groups(A , B and C)randomly . First , we mensurated each parameter of 3 groups . Second , patients in group A were taken placebo ; patients in group B were taken huidi(HD, Mesalazine enter-coated tablets) 1g each time , three times a day , moreover 1g HD was used as retaining enema which melted into 20ml 2% lidocaine and 150ml physiological saline every night ; patients in group C were taken 100ml KJT each time , two times a day , moreover 150ml KJT and 20ml 2% lidocaine was used as retaining enema every night . Finally , each parameter of 3 groups was mensurat-ed again when the patients were cured for 6 weeks . 3.Metricalparameter: (1)Patients'age , sex , course of disease before cur-ing (.2)Clinical grade . (3)Grade of colonoscope . (4) Grade of pathology . (5)NF-κB p65 immunohistochemistry staining , which were analyzed by image analysis system . Result: 1.Each parameter and test result before curing: Difference of each parameter of 3 groups had no statistical significance by one-way ANOVA test , chi-square test and rank sum test(P>0.05). 2. Change of each parameter of each group and test result after cuing: Difference of each parameter of group A after curing had no statistical significance by rank sum test compared with that before curing (P>0.05); difference of each parameter of group B after curing had statistical significance by rank sum test compared with that before curing(P<0.05); difference of each parameter of group C after curing had statistical significance by rank sum test compared with that before curing(P<0.05). 3. Change of each parameter between 3 groups and test result after cuing: Difference of each parameter between 3 groups had statistical significance by rank sum test (P<0.05). 4.Grade of pathology , NF-κB positive reactions cells'area density and amount density had positive correlation relationship by Spearman rank test(P<0.05). Conclusion: 1.It is effective to treat patients of UC with KJT , and the curative effect of KJT is better than that of HD .
