Atherosclerosis is a multifactorial disease. Its risk factors are mainly composed of age, sex, blood pressure, blood lipids and blood glucose and so on. Recently, the relationship between blood uric acid (UA) and atherosclerosis was observed, but the mechanism of which is not absolutly clear. In this research, the risk factors of atherosclerosis were observed , relationships among the factors and mechanisms were explored by cell culture technique, ELISA and epidemiologic analysis.Experiment One: Effect of uric acid on proliferation of rat vascular smooth muscle cells.Objective: To observe the effect of uric acid on proliferation of rat vascular smooth muscle cells, the key step of atherosclerosis. Methodes: Primary cultured vascular smooth muscle cells (VSMC) were used and divided into control group, Uric acid (UA) group (2 mg/dl, 4 mg/dl, 6 mg/dl) and UA & losartan group. UA (4mg/dl) group was further divided into three sub-groups by different incubational time (24, 48, 72 hours). MTT assay and flow cytometry (FCM) were used to detect the proliferation of the cells. Results: UA significently increased the values of MTT absorbance (4mg/dl UA:0. 65 ± 0. 06, P<0. 01 conpared with control = in a time-and dose-dependent manner. This effect could be partly blocked by losartan (0.59±0.05, P<0.01 conpared with control). FCM showed that the percentage of cells in Gl phase decreased and the percentage of cells in S phase increased after UA treatment. Conclusion: UA can accelerate the proliferation of VSMC in a time-and dose-dependent manner, and the receptors of angiotensin II may play a role in the effects of UA stimulated VSMC proliferation. Experiment Two: Clinical analysis of risk factors of atherosclerosis in type 2 diabetes mellitus and their relationships.Objective: To investigate risk factors of carotid atherosclerosis in type 2 diabetes mellitus and its relationsships.Methodes: 176 T2DM patients with or without carotidatherosclerosis were studied in case-control method. Age, sex, systolic blood pressure(SBP), diastolic blood pressure(DBP), fasting blood glycose(FBG), waist hip rate(WHR), serum cholesterol (TC), triglyceride(TG), low-density lipoprotein cholesterol(LDL-c), UA and IGF-1 were analyzed. According to a modified WHO definition for hypertension, T2DM patients with or without carotid atherosclerosis are classified into two groups respectively.Results: The levels of age, UA and IGF-1 were significantly increased in T2DM patients with carotid atherosclerosis. UA was increased in T2DM patients with carotid atherosclerosis and hypertension and T2DM patients with hypertension, while the result of IGF-1 was different. With correlation analysis, UA positively correlated with LDL and IGF-1. With multiple logistic regression, age, LDL, UA, IGF-1 were independently associated with carotid atherosclerosis.
