Objective To observe the expression of nAchR and NGF in the brain of ovarie-ctomized rats with transection of fimbria/fornix /estrogen treatment and understand their effects on AD.Method Healthy female wistar rats were randomly divided into 5 groups: (1) control group;(2) fimbria/fornix transection and ovariectomized group;(3) ovariectomized group;(4) fimbria/fornix transection group; (5) estrogen treatment group; There were five rats in every group. And fimbria-fornix of brain was transected and ovary was cut off in the stereotaxic apparatus in order to make AD model. Immunoreactive cell of nAchR and NGF were stained by immunohistochemical way. To observe the expression varieties of nAchR and NGF immunoreactive cell in the cortex, hippocampus CA1, amygdale and Meynert nucleus.Results The expression of nAchR and NGF did not distinctively change in the cortex, hippocampus CA1, Meynert nucleus, amygdale in control group. While in fimbria/fornix transection of group, a number of cholinergic neuron undergoing lysis,the expression of nAchR remarkably decreased (P<0.05) .And in ovariectomized group, the level of estrogen in the plasm decreasing ,the expression of nAchR obviously decreased in the observed areas of the brain (P<0.05 ) .And in fimbria/fornix transection and ovariectomized group, the expression of nAchR occured the same changes . And the expression of NGF did not distinctively change in the observed areas of cortex, hippocampus CA1, amygdale of the brain of every AD model.But in Meynert nucleus, the expression of NGF remarkably decresed than control group.In estrogen treatment group ,the expression of nAchR increased.and NGF did not change .Conclusion Fimbria/fornix was transected and ovary was cut off in order to simulate AD model and injected rats with estrogen , then to look into the pathological changes in AD brain. Founds testified that the expression of nAchR decreased and the expression of NGF did not change in the areas of cortex, hippocampus CA1, amygdale of the brain in AD model groups. But in Meynert nucleus, the expression of NGF remarkably decresed than control group.Injection of estrogen increased the expression of nAchR . So...
