Objective To observe the expressive variety of brain-derived neurotrophic factor (BDNF) in the different areas of rats with the model of bilateral fimbria/fornix transection and ovariectomization according to 2 x 2 ANOVA design, the expressive variety of BDNF was observed in the model rats with the estrogen replacement therapy.Methods Healthy female rats were randomly divided into 4 groups according to 2 x 2 ANOVA design, there were five rats in every group: (1)group(OVX group); (2) fimbria/fornix transection of group(FF group); (3)fimbria/fornix transection and ovariectomized group(OF group); (4)control groups. In the treatment parts: Healthy female rats were randomly divided into 2 groups: (1) fimbria/fomix transection and ovariectomized group(OF group); (2)estrogen replacemen therapy group(E2 group). The bilateral fimbria-fornix of brain were transected in the stereotaxic apparatus, and bilateral ovaries were ablated in order to erect cognition disorder model. Immunoreactive cells of BDNF were stained by immunohistochemical way. To observe the expressive varieties of BDNF immunoreactive cell in the hippocampus CA1, cerebral cortex, almond complex and Meynert nucleus. The number of masculine cell were noted in high times microscope at random selected 5 fields in every detected area, and then compared the average number of BDNF according to 2×2 ANOVA analysis amoung the fuor groups. In the treatment parts, the average number of BDNF between OF group and E2 group was compared with SPSS analysis.Results The bilateral fimbria/fornix transection and ovariectomization had a interaction on the expression of BDNF in every brain areas(P<0.01). The expression of BDNF remarkably decreased in hippocampus CA1, cerebral cortex, almond complex and Meynert nucleus of the brain in the model groups with the bilateral fimbria/fornix transection and ovariectomization(P<0.01), and the expression of BDNF in Meynert nucleus area of the OF group was decreased remarkably than the other...
