Objective: To investigate the expression of mRNA and protein of vascular endothelial growth factor (VEGF), its receptor KDR and their relationship to metastasis and prognosis in pulmonary carcinoma.Methods: The expression of mRNA and protein of VEGF and KDR was examined differently by in situ hybridazation(ISH) in 30 fresh non-small-cell-lung- cancer (NSCLC) samples and PV-9000 method ,a kind of immunohistochemistry (IHC) in 75 patients with pulmonary carcinoma.Results: 1, The distribution of VEGF and KDR was identical, mainly located in the cytoplasm of tumor cells (TC), cytoplasm of fibroblasts (FB),endothelial cells (EC) and less in the cytoplasm of inflammatory cells .The positive rate of VEGF in tumor cells were61.33% (46/75 ) by IHC and66.67% (20/30) by ISH, and the two rates showed no significant difference (x~2=0.261,P>0.05) .The positive rate of KDR in tumor cells were76.00% (57/75 ) by IHC and73.33% (22/30) by ISH, and the two rates also showed no significant difference (x~2=0.082, P>0.05).2 , There were significantly positive correlations both between the positive rate of KDR and VEGF detected by IHC and ISH.( r= 0.768,0.853; respectively P <0.05).3, No significance was found between the expression of VEGF or KDR protein in age groups, gender groups , pathological type and pathological grade groups in the TC (x~2=0.04~3.06,P>0.05); Also no significance was found between the expression of VEGF or KDR mRNA in pathological type and pathological grade groups in the TC(P=0.419~1.000).4, The positive rate of VEGF and KDR protein in TC, in D>5cm group, 3cm
