Expression of vascular endothelial growth factor receptors (VEGFRs) in pulmonary carcinoma and their relationship to metastasis of lymph node and prognosisObjective: To investigate the expression of three kinds of vascular endothelial growth factor receptors (VEGFRs)-Fltl, KDR, Flt4 and their relationship to metastasis and prognosis in pulmonary carcinoma. Methods: The expression of Fltl, KDR and Flt4 was detected immunohistochemistrily by PicTure?PV9000 method in 75 patients with pulmonary carcinoma. Results: 1.There was an extensive expression of Fltl and KDR, mainly in the cytoplasm and plasmalemma of tumor cells (TC), cytoplasm of fibroblasts (FB), endothelial cells (EC) and in inflammatory cells. Flt4 was mainly existed in TC cytoplasm, and was less in FB. They also could be found in nucleus of small cell lung carcinoma (SCLC). 2.The positive rate of Fltl, KDR and Flt4 was 65.33%, 76%, 69.33% in TC; 45.33%, 57.33%, 37.33% in FB. The positive rate was much higher in TC thaninFB(x2=6.068, 5.88, 15.429; P=0.017, 0.019, 0.00l).3.In both TC andFB.no significance was found between the expression of Fltl,KDR and Flt4 in age groups ( x 2 =0.537, 1.919, 3.335; 1.072, 0.71, 2.938;P=0.765,0.383, 0.189; 0.585, 0.701, 0.23).In gender groups, the difference of positive rate of each receptor was of no significance ( x2 =1.78, 1.292, 0.742; 0.013, 0.668, 0.698; P=0.182, 0.256, 0.389; 0.908, 0.414, 0.403 ) .Difference also could not be found in groups of pathological types (P=0.374,0.287,0.24,0.356,0.786,0.312) or histological grades( x 2=0.457,3.234,3.464; 4.844,1.573,0.762;P2=0.796,0.199,0.177;0.089,0.455,0.683). 4. In TC and FB, the positive rate of Fltl, KDR and Flt4 in D^Scm group was 81%,88%,84%,56%, 59%,44%. In 3cm5 group was 36%,48%,44%,24%,28%,24%. The positive rate in TC was significantly different in the three groups(x2=17.87, 16.19, 14.03; P=0.000,0.000,0.001),but it wasn't of same significance in FB(x2=8.812,19.19,5.094;P=0.012,0.000,0.078). 7.In TC the expression had a striking positive correlation between each pair of the three kinds of receptors(Flt1 andKDR, Flt1 andFlt4,KDRand Flt4;r=0.443,0.366,0.303; P=0.0004, 0.0036, 0.009) . Conclusion: l.Our findings suggest that the three receptors mainly express in TC, and have expressive synergistic effect in TC. l.The expression of Fltl, KDR and Flt4 in TC is related to tumor size , lymph node metastasis and the postoperative survival time of the patient. The same relation in FB also be found in Fltl and KDR with tumor size or lymph node metastasis. 3.The positive rate of all the three receptors both in TC and FB has no difference in age, gender groups, pathological types and histopathological gradings. 4.Detecting of the expression of Flt1, KDR and Flt4 may be more valuable for evaluating the metastasis and prognosis of pulmonary carcinoma.Postgraduate: Ma Guixin(Department of Pathology) Directed by Xiang Fenggang...
