Objective: To investigate the roles of Th1 and Th2 cell reaction in the pathogenesis of EAN, an animal model of Guillain-Barre syndrome (GBS).Methods: (1) Eighteen New Zealand white rabbit) were divided into three group(FKG group, FK group and FG group). GM1, FCA and KLH were combined to FKG, FK and FG mixture respectively and used to immunize rabbits to make EAN. Immunization was made for three times. The interval time of immunization was in every 20 days. Clinical changes of EAN and blood samples were collected every 10 days. (3)Titers of GM1 antibodies were determined with ELISA methods and mRNA levels of IFN and IL-10 were determined with QIAGEN one step RT-PCR kit. The data were analyzed by using one way analysis of variance and T test. P<0.05 means that there was difference in statistics. P<0.01 means that there was significant difference in statistics.Result: Temporal decreased muscle strength was noted. GM1 IgG antibody began to rise after immunization and reach the climax after 40 days of immunization. The expression of IFN- γ mRNA increased gradually after first immunization, and reached the highest level at the 40 days after first immunization, and decreased after the climax. There was no difference between FKG and FK group. But there was significant difference between the former two groups and FG group. The expression of IL-10 mRNA began to increase gradually at 30~40 days after first immunization. There was no difference between FKG and FK group in early stage of immunization. There was difference between the former group and FG group. At the later stage of the immunization, there was difference between FKG and FK.Conclusion: Both Thl and Th2 reactions play important role in the pathogenesis of EAN induced by immunization of GM1. Protein carrier may induce both Thl and Th2 reactions. Polysarccraid antigens may also induce Th2 reaction. Protein carrier play an important role in the production of GM1 antibody.
