Objective Thrombosis is a significant factor in the occurrence of many cardiovascular diseases and does great harm to human health. The timely and accurate diagnosis of thrombotic events and the therapy effect of the thrombolysis play a key role in the prognosis of these disorders. The tissue-type plasminogen activator(t-PA) played a significant role in fibrinolytic system.,whose activity abnormity is the most hazardous factor in the prethrombotic state of cardiovascular disease. We studied the mRNA expression of t-PA in the peripheral blood nucleated cells from the thrombosis group and prethrombotis group by using RT-PCR and investigated the difference between the patients and healthy individuals.Methods All peripheral blood samples were obtained from patients in nerologic and cardiovascular wards of Yantai Yuhuangding Hospital from October to December in 2004. 40 patients were diagnosed as cerebrol infarction, 20 as primary hypertension, 20 as coronary artherosclerotic heart disease, and 40 healthy persons served as control. The t-PA mRNA in peripheral blood nucleated cells was detected by RT-PCR and the t-PA activity was determined by chromogenic substrate method.Result The mRNA expression levels of t-PA from peripheral blood nucleated cells in the thrombosis group and pro-thrombosis group were remarkable high and notable difference was shown comparing with those of the normal group(P<0.01 ).The activities of plasm t-PA in the former two groups were reduced distinctly and had notable difference (P<0.01 ) .The mRNA expression of t-PA from peripheral blood nucleated cell had a close correlation with prethrombotic state. The experimental results were shown using mean value ±standard deviation.analysed by using statistical soft ware Sigmastat 2.0.The statistical methods being used were listed as t-test and regress-analyse, while P<0.05 and P<0.01 means that the differences had statistical senses and notable statistical senses.Conclusion The mRNA expression of t-PA in the peripheral blood nucleated cells is closely correlated with the thrombosis and prethrombotis. which may be an early assistant indicator of the prethrombotic state. |
