Objective:To study the expressions and significance of Survivin mRNA in tumor tissues and blood of patients with renal dear cell carcinoma(RCCC).Methods:Survivin mRNA in tumor tissues of 46 patients with RCCC and 15 normal kidney tissues were detected by RT-PCR.RT-PCR-ELISA was used for testing the Survivin mRNA expressions in blood of 46 patients with RCCC,and 15 healthy volunteers.Results:1.The positive expression rate of Survivin mRNA in tumor tissues was 63.0% (29/46),which was notably higher than the 6.7%(29/46) expression rate in normal kidney tissues.The positive expression rates in clinical stages ofⅠ,Ⅱ,ⅢandⅣwere 36.4%,50.0%,76.9%and 100%respectively,and in pathological grades of G1,G2 and G3 were 36.3%,90.5%and 100%respectively.The expression level of Survivin mRNA was markly elevated as the clinical stage and pathological grade increased in RCCC. (p<0.05).The positive expression rate of Survivin mRNA in patients with lymphatic metastasis was 100%,while was 45.2%in patients without lymphatic metastasis,which showed significant difference with relationship to lymphatic metastasis(p<0.01).The expression rates of Survivin mRNA were not significantly different in associated with gender,age and the size of the tumor(p>0.05).2.21 of 46(45.7%) showed positive Survivin mRNA signals in blood of patients with RCCC.While only 2 of 15(13.3%) patients in control group showed positive Survivin mRNA signals.The expression rates of Survivin mRNA were significantly different between experimental group and control group(p<0.05).The sensitivity and the specificity of the expression of Survivin mRNA in peripheral blood were 45.7%and 86.7%,respectively.There was no significant correlation with clinical stage,pathological grade and other clinical features.3.The positive expression rates of Survivin mRNA were 63.0%in tumor tissues and 45.7%in blood respectively,Which indicated correlationship between the two specimens(p<0.01).Conclusions:1.Tumor tissues of patients with RCCC show significant positive expression of Survivin mRNA,and the expression is correlated with clinical stage, pathological grade of RCCC.The up-regulated expression of Survivin mRNA in RCCC suggests that Survivin mRNA is tighlty related with the tumor's occurrence and invasion. Therefore,Survivin mRNA might be a new index for identifying the prognosis of RCCC.2.The application of RT-PCR-ELISA to detect the expression of Survivin mRNA in blood shows a high sensitivity and specificity and a minimal invasion.Thus, Survivin mRNA might be a new index for RCCC diagnosis.
