Objective To observe the effects of ginkgo biloba extracts(EGb761) on eiythrocyte lipid peroxidative injury and erythrocyte survival, and to explore the protective mechanism of EGb761 on erythrocyte.Methods (1)Iipid peroxidative injury: Intravenous blood samples taken from healthy adults were prepared for red blood cell(RBC) suspensions and divided equally into 6 groups: group I for normal, group II for control, with hydrogen peroxide (H2O2, 100mmol/L) induced injury, and group III, IV, V, VI with the same injury as group II , but being pretreated with different concentrations of EGb761 (0.002g/L, 0.010g/L, 0.050g/L, 0.250g/L), respectively.(2)Senescent erythrocyte: Intravenous blood samples taken from healthy adults were divided equally into 5 groups: group I for control, group II, III, IV, V were respectively pretreated with different concentrations of EGb761, incubated under 37℃ for 12h, 24h and 36h as erythrocyte normal aging. Hemolytic degree, the concentration of erythrocyte membrane lipid peroxidation malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) and ATPase in RBC suspensions after incubation were measured. At the same time, membrane skeleton structure and morphology of erythrocyte were studied with transmission electron microscope(TEM) and scan electron microscope(SEM) for erythrocytes of lipid peroxidative injury, respectively.Results After lipid peroxidative injury, the MDA concentration increased significantly while the activity of SOD and Na+, K+-ATPase and Ca2+, Mg2+-ATPase decreased significantly compared with the normal(t = 2.02 70.06, p < 0.05, 0.001).After the addition of EGb761, the hemolytic degree decreased significantly(t = 4.13 19.41, p<0.05, 0.001), the MDA concentration also decreased significantly while the activity of SOD and Na+, KV+-ATPase and Ca2+, Mg2+-ATPase increased significantly compared with the control group (t = 2.02 16.88, p < 0.05, 0.001).Significance could also be seen within the different EGb761 concentration groups(q = 3.14 83.89, p < 0.05,0.01 ).After normal aging, the MDA concentration increased while the activity of SOD and ATPase decreased significantly compared with the normal(t = 2.02- 74.99, p<0.05, 0.001) .With the addition of EGb761, the concentration of MDA decreased and the activity of SOD and ATPase increased significantly, compared with the control(t = 2.02 75.11, p < 0.05,0.001). Significance could also be seen within the groups pretreated with different concentrations of EGb761(F = 214.37 1591.52, q = 5.72 100.85, p < 0.05, 0.01). The erythrocyte membrane skeleton structure of groups injured was loose, blurred and partially deficient. Groups pretreated with EGb761 became better. Obviously,...
