Solanum nigrum polysaccharide from solanaceae Solanum nigrum and tomato genus Solanum nigrum L. all the grass extract of bioactive macromolecular composition,in recent years the pharmacological actions have more study reports,especially in its clinical application and pharmacological mechanisms have more found in antitumor,and there is also a greater progress. Through reports in the literature,found a tumor-burdened mice red cell membrane liquidity,closed degrees and saliva acid and super oxide content of enzyme guhya than normal mice red cell membrane lower. Otherwise reports in the literature solanum nigrum polysaccharides by enhancing immune function to red blood cells in mice tumor-burdened anti-tumor effect,show solanum nigrum polysaccharide on tumor have certain effect,and the role and red blood cells have the certain relation,but of solanum nigrum polysaccharide regulation tumor-burdened mice red cell membrane function effects not were reported,therefore this paper red cell membrane function influence perspective of solanum nigrum polysaccharide antitumor function of research.Transplanted tumor cells by in vivo modeling methods were Solanum nigrum polysaccharide Pharmacodynamic Study; labeled with fluorescent probe DPH fluorescence spectrophotometer S180 and H22 tumor-bearing mice with the red cell membrane fluidity,the red cell membrane liquidity size can use fluorescence polarization degree,micro viscosity and liquidity to say,By colorimetric method for determining the red cell membrane shadow bubble in add surfactant and do not add surfactant when the enzyme activity changes to the detection of solanum nigrum had S180 polysaccharids H22 tumor-burdened mice and red cell membrane closed degrees influence; Adopt kit S180 and determination of solanum nigrum polysaccharide H22 tumor-burdened mice red cell membrane saliva acid content and RBC superoxide dismutase (SOD) content of influence of research,to further research and polysaccharides solanum nigrum clinical application provides important experiment basis.Solanum nigrum polysaccharide was researched on the pharmacodynamics. pharmacodynamics first established in mice S180 sarcoma model and H22 ascites tumor model respectively,the S180 and H22 mice inhibition rate and prolonged life rate were found.solanum nigrum polysaccharide 31.25 mg/kg,62.50 mg/kg,125mg/kg doses groups all can inhibit S180 solid tumor growth,its average tumor weighs negatie were significantly lower than those in control group(P<0.01),including solanum nigrum polysaccharide 125mg/kg doses group inhibition rate reach 55.62%. Explain solanum nigrum polysaccharide on S180 solid tumor has good inhibitory effect. The results also show that solanum nigrum polysaccharide 31.25 mg/kg,62.50 mg/kg,125mg/kg dose group of H22 mice can prolong surial,solanum nigrum polysaccharide 62.50 mg/kg,125mg/kg dose group of life extension rate were achieved respectively 55.81% and 65.12%,and compared with negative group,difference is very significant(P<0.01).By SNL on S180 and H22 tumor on erythrocyte membrane fluidity were studied, respectively S180 and H22 tumor-bearing mice were measured membrane fluidity was found SNL can significantly improve the cell membrane S180 tumor mice mobility.125mg/kg dose group, which increased S180 tumor cell membrane fluidity in mice was 5.1618±0.0763, SNL 62.50mg/kg dose increased S180 tumor cell membrane fluidity in mice was 4.9329±0.0430, compared with the negative control The difference was significant (P<0.01). That SNL can significantly improve the H22 tumor mice membrane fluidity. H22125mg/kg dose group, which increased tumor cell membrane fluidity in mice was 5.3092±0.4801, with the negative control group, the difference was significant (P<0.01). SNL 62.50mg/kg dose of H22 tumor mice increased membrane fluidity was 4.8243±0.4152, compared with the negative control group, significant difference (P<0.05).by SNL on S180 and H22 tumor bearing mice and blocking degree of red cell membrane, and found that SNL 62.50mg/kg dose group and 125mg/kg dose group could increase the S180 tumor-bearing mice blocking degree of red cell membrane, respectively, was 37.73±4.24% and 55.01±3.85%, compared with the negative control group, significant difference (P<0.05, P<0.01). SNL can significantly improve the H22 tumor-bearing mice blocking degree of red cell membrane, where SNL 62.50mg/kg dose group and the 125mg/kg dose group H22 tumor-bearing mice can enhance blocking degree of red cell membrane closure was 29.22±3.70%, and 30.52±1.41% respectively, compared with the negative control group, significant difference (P<0.05).Through solanum nigrum polysaccharids on S180 and H22 tumor-burdened mice red cell membrane saliva acid content impact study,with kit for red cell membrane on mice tumor-burdened determination of SA content,the results showed that the solanum nigrum polysaccharide 62.50mg/kg,125mg/kg doses groups can significantly improve the SA content,and compared with negative control group,difference is very significant (P<0.01),and solanum nigrum polysaccharide 125mg/kg dose group of S180 and H22 mice of SA content achieved 1.9545±0.0624mmol/gprot,0.1620±0.0140mmol/gprot, respectively. Solanum nigrum polysaccharide 62.50mg/kg S180 and H22 dose group of mice SA content were 2.0227±0.2714 mmol/gprot,0.1146±0.0203mmol/gprot, and compared with negative control group,there exists significant difference (P< 0.05, P<0.01).In S180 and H22 tumor-burdened mice RBC SOD content affected experiment that: solanum nigrum polysaccharide 62.50mg/kg and 125mg/kg doses group were significantly improve tumor-burdened mice erythrocytes are superoxide dismutase (SOD) content,and 62.50mg/kg polysaccharide of solanum nigrum dose group of H22 and S180 mice SOD reached 13.44±2.16U/mgHb,5.11±3.89U/mgHb, and compered with negative control group, respectively,difference (P<0.01,P<0.05). Solanum nigrum polysaccharide 125mg/kg dose group of S180 and H22 mice SOD activity reached 13.70±0.90U/mgHb,5.74±0.66U/mgHb, respectively,and compared with negative control group,significant difference (P<0.01,P<0.05). Solanum nigrum polysaccharide of 31.25mg/kg doses groups can achieve S180 mice SOD activity 12.56±1.30U/mgHb,compared with negative control group,significant difference (P<0.05).To sum up,solanum nigrum polysaccharids significantly inhibit portability S180 tumor cells to grow,to transplant H22 has obvious prolonged life function; Meanwhile,solanum nigrum polysaccharides could evidently increase tumor-burdened mice red cell membrane liquidity and closed degrees,improve tumor-burdened mice red cell membrane saliva acid content and RBC superoxide dismutase (SOD),the results showed that the content of solanum nigrum polysaccharides by improving and recovery may tumor-burdened mice red cell membrane function,thus achieved the antitumor function.
