Objective: To provide scientific experimental bases for clinic treatment of kidney weak osteoporosis by observing the effects that nanometer calcium kidney-tonifying herbs have on the BMP-4, Smad6 mRNA and protein expression of the hypothalamus of rats with kidney weak osteoporosis, exploring the molecular biology bases of Chinese herbal prophylaxis and treatment to tonify the kidney during the disease time of osteoporosis, and researching the theories how nanometer calcium kidney-tonifying herbs prevent and treat the rats with kidney weak osteoporosis.Method: Build a model of kidney weak osteoporosis by removing two-sided ovaries. Treat the rats with different (high, medium and low) dosage of nanometer calcium kidney-tonifying herbs for 12 weeks (applying reduction to absurdity), while taking Gushukang Keli and oyster calcium as positive controls, and also set normal control and wild control. Mensurate the skeletal density of the left femur of the rats with Model XR-36 double energy X-ray skeletal density detector (US NORLAND). Detect the indexes of TRAP and ALP of the serum with biochemical methods. Detect the BMP-4, Smad6 mRNA and protein expression of the hypothalamus of rats with kidney weak osteoporosis by using RT-PCR and Western imprinting. Results:Conlusions: 1) Comparison of the hyster indexes of experimental animals: The indexes of the wild control decreased obviously compared to the normal control. That of the nanometer calcium control, Gushukang Keli control and oyster calcium control increased slightly compared to the wild control, but there is no prominent difference. 2) BMD data of the skeletal density of the left femur of experimental animals: All the medicines (nanometer calcium kidney-tonifying herbs, Gushukang Keli and oyster calcium) can increase the femur skeletal density of the rats whose ovaries were removed greatly after 12 weeks, while the control with nanometer calcium kidney-tonifying herbs has the best effects. 3) TRAP - biochemical index of serum osseous metabolism: Compared to the wild control, the nanometer calcium kidney-tonifying herbs controls with low and medium dosage have notable change, butnot reach the level of the normal control. When compared to the normal control, distinct differences exist in all the controls, but there is no obvious difference among these controls. 4) Content of serum ALP: The nanometer calcium kidney-tonifying herbs controls with low and medium dosage are notably different from the wild control, and they can reach the level of the normal control. 5) BMP-4 mRNA and protein expression of the hypothalamus of rats detected by RT-PCR and Western imprinting: BMP-4 mRNA and protein expression of the hypothalamus of rats in the wild control increased obviously compared to the normal control. 6) Smad6 mRNA and protein expression of the hypothalamus of rats detected by RT-PCR and Western imprinting: Smad6 mRNA and protein expression of the hypothalamus of rats in the wild control decreased notably compared to the normal control.Conclusion: Nanometer calcium kidney-tonifying herbs can distinctly increase the skeletal density and done formation of the rats with osteoporosis and restrain skeletal absorption. BMP-4 mRNA and protein expression of the hypothalamus of rats in the wild control increased obviously compared to the normal control, while Smad6, mRNA and protein expression of the hypothalamus of rats in the wild control decreased notably compared to the normal control.
