Abstract/Summary:
Objective: nm23-H1 is a candidate gene for the suppression ofcancer metastasis. Several studies on human breast, hepatocellular,gastric, ovarian and melanomas have shown that reducednm23-H1 expression was closely related to metastatic progressionwith poor prognosis; matrix metalloproteinase-2 and tissueinhibitors of metalloproteinases-2 control the breakdown ofextracellular matrix components and play an important role in theprocess of invasion and metastasis. Thus, to investigate therelationship of the expressions of nm23-H1,MMP-2 and Timp-2in bladder carcinoma have very important significance. Methods:Immunohistochemical study for nm23-H1,MMP-2and Timp-2 were performed on 69 samples of bladder transitionalcell carcinoma of paraffin-embedded specimens of surgicalcarcinoma. Results:There were significant difference in nm23-H1 andMMP-2 between the bladder transitional cell carcinoma and thenormal bladder epithelium(P<0.01). The positive expression ofthe nm23-H1 was decreased with the stage ,grade and metastasis(P<0.05). But the positive expression of the MMP-2 wassignificantly increased with stage and grade ( P <0.05 ) .Furthermore, the positive rates of the MMP-2 was increased intumor diameter greater than 2cm, which showed statisticalsignificance(P<0.05). The positive expression of the Timp-2 wasdecreased with the stage and grade and the expression intensity ofthe Timp-2 was increased with stage and grade, but which didn'tshow statistical significance. Conclusion: The expression of some metastasis-related genescan be used to estimite the metastatic potenciality and help for theadguvant treatment.
