Objective To observe influence of culture system on colony formation, study effect of psoriatic peripheral blood mononuclear cells (PBMCs) on activity of marrow high proliferative patential colony forming cell(HPP-CFCs) and reveal the relation of immunocytes and HPP-CFCs from psoriatic patients. Methods Marrow mononuclear cells were separated by density gradient centrifugalization and cultivated in methylcellulose semi-solid culture medium with cytokines and PHA-stimulated condition to establish idealer culture system for HPP-CFCs . The HPP-CFCs were cultured in the culture system stimulated respectively with PBMCs culture supernatant derived from psoriatic patients and normal controls, and the colony number of HPP-CFCs was counted on the 14th day after culture. Results HPP-CFC colonies formed in medium both with PHA-MCM and with cytokines, however the colonies were more in cytokines system than in PHA-MCM one(P<0. 05) . Compared respectively with the group of spontaneous proliferation and the group stimulated by PBMCs culture supernatant from normal controls, the HPP-CFC colonies decreased significantly (P<0.05); However, there was no statistical significance between the group stimulated by PBMCs culture supernatant from normal controls and the spontaneous proliferation group(P>0.05). Conclusion The cytokines culture system is superior to the PHA-MCM one; Colony formation of HPP-CFCs can be suppressed by psoriatic PBMCs culture supernatant, which suggest that psoriatic PBMCs have specific biological activity; The marrow microenvironment can be affected by peripheral blood environment , so the activity of psoriatic patients HSCs is probably abnormal.
