This study has three parts: In the first part we constructed recombinant eukaryotic expression vector pIRES-CIITAm and recombinant adenoviruses Ad-CIITAm both containing CHTA mutant gene and recombinant adenoviruses Ad-CIITAm-pIV containing CIITA mutant gene which is regulated by pIV. In the second part we constructed a recombinant adenoviruses Ad-FasL containing FasL gene and the other recombinant adenoviruses Ad-FasL-pIV containing FasL gene which is regulated by pIV.The results of in vitro tests in the third part demonstrated CIITA mutant gene transfer mediated by pIRES-CIITAm and Ad-CIITAm could inhibite inducible expression of class II MHC molecules,and tentatively confirmed that FasL gene transfer mediated by Ad-FasL could induce apoptosis of cells with Fas receptors on it.
