The paper is designed to study the anti-tumor activity of Ethanol Extract of Fomes fomentarius(L.:Fr.)Kick. (EEF) and its possible Mechanisms.The effects of EEF on tumor growth, survival days, immune function and the expression of apoptosis-associated genes bax, bcl-2 of tumor-bearing mice were determined through S180 tumor model.The effects of EEF on MDA contents and primary antioxidant enzymes activities of normal mice were measured.The influences of EEF on Hela, S180 and normal spleen cells were assayed using MTT method. The apoptosis rate of S180 cells incubated with EEF was analysed by FCM. In addition, the effects of EEF on S180 and normal spleen cells were observed through serum pharmacological method. The results were shown as follows. Firstly: 100mg/kg.d, 250mg/kg.d, 500mg/kg.d EEF were found to remarkably inhibit tumor growth, prolong survival days of mice carrying S180 tumor, enhance cellular immunity, humoral immunity, non-specific immunity , bax gene expression and restrain bcl-2 gene expression of tumor-bearing mice at the same time . Secondly: MDA contents of liver and kindey were decreased in 250mg/kg.d, 500mg/kg.d groups; SOD activity of liver and kidney , GSH-PX activity of liver in normal mice were advanced in 100mg/kg.d, 250mg/kg.d groups, respectively. Thirdly: obvious proliferation-inhibitory activities to Hela, S180 cells in vitro were found in 31.25500μg/ml groups; and also the growth of normal mouse splenocytes was enhanced in 125μg/ml group. Fourthly: The apoptosis rate , cells at G0/G1 phase were obviously increased and cells at G2/M and S phases were remarkably decreased, after S180 cell were dealt with 100μg/ml,400μg/ml EEF. Fifthly: the growth of S180 cells treated with serum containing EEF was significantly inhibited and spleen proliferation was promoted by serum containing EEF. EEF has obvious anti-tumor effect to S180 tumor. The anti-tumor mechanisms of EEF can be connected with the enhancement of immune function and antioxidant activity, the high directive cytotoxicity to tumor cells, the promotion to tumor cells apoptosis rate, the blocking to cell cycle of tumor cells.
