This thesis studied the chemical constituents,the anti-tumor and antimicrobial activities of Fomes fomentarius body and the pharmacological activities of their fermentation products..In the aspect of chemical composition research, two compounds were separated from the petroleum ether extract and the chloroform extract of Fomes fomentarius by the refluxing technique, thin layer chromatography and silica gel column chromatography. The compounds were identified as [4,6,8(14),22(23)-tetraen-3-one-ergostane]and[7(8) 22(23)-diene-3-one-ergostane] through the MS,1H-NMR,13C-NMR and DEPT.One compound separated from the methanol extract of Fomes fomentarius body using the HPLC technique was identified as [3,3-dimethyl-(8-hydroxy-3--octenyl) cyclohexanol] through'H-NMR,13C-NMR and DEPT.In the aspect of anti-tumor research, the supernatant and the precipitates of petroleum ether extract, ergosterol and ergosterol peroxide,the chloroform extract,and the methanol extract of Fomes fomentarius body were applied to H22 tumor-bearing mice for the anti-tumor experiment. The results showed that supernatant and precipitates of petroleum ether extract, ergosterol and ergosterol peroxide, methanol extract of Fomes fomentarius body can inhibited the growth of tumor. Compared with positive drug control group, the spleen index and the thymus index were observed to be obviously increased, as well as the H22's leukocyte number and the serum IL-2 content of H22 tumor-bearing mice all increased obviously too.It demonsteated that the petroleum ether extract, ergosterol and ergosterol peroxide of Fomes fomentarius body inhibited tumor growth, and thereby enhancing the immunity of organism. In addition, ergosterol and ergosterol peroxide were induced apoptosis in human hepatoma cell experiments, the results show that ergosterol and ergosterol peroxide could induce apoptosis of hepatoma cells, to inhibit the effect of liver cell division, Further treatment of cancer diseases.In the study of antimicrobial activity, an in vitro antimicrobial experiment was conducted using the methanol extracts of Fomes fomentarius body. The result indicated that not only the obvious inhibitory action to E.Coli ATCC8099 and staphylococcus aurous ATCC6538 it has, but also the antimicrobial activity has relationship with the dose-effect. When the concentration of the methanol extract was 50 mg/ml, the antimicrobial rates of E.Coli ATCC8099 and staphylococcus aurous ATCC6538 reached 99.39%和95.10%,respectively.In the aspect of fermentation studies, a liquid rocking bed culture of Fomes fomentarius body was performed, and the formed fermentation products was used in vitro antimicrobial experiments and in vivo anti-tumor experiments. In the former case,it was observed that the products of Fomes fomentarius fermentation contained similar antimicrobial-active constituents as the ones in Fomes fomentarius body.Moreover,the bacteria solution exhibited higher antimicrobial efficiency than colonies.The results indicated that the products of Fomes fomentarius fermentation had a good pharmacological activity.Further studies and development would be performed.In addition,the Fomes fomentarius fermentation exhibited a short production time and a high yield,which made a large-scale production possible in the future.The accomplishment of the work would congtribute to the development and utilizationg of fungal drugs.In the in vivo anti-tumor experiments,the results showed that the Fomes fomentarius fermentation products had a pronounced inhibitory effect on the tumor growth of H22 tumor-bearing mice, and it has relationship with the dose-effect,indicating that the fermentation products might also contain similar anti-tumor active constituents as the ones in Fomes fomentarius body. Further investigations were under the process.
