The Expression Of Survivin In The Breast Cancer And Its Correlation With The Expression Of P53 Gene

purpose :To detected the expression of survivin in normal breast tissue , simple hyperplasia,laryngeal atypical hyperplasia and breast carcinoma by using ABC immunohisto-chemical method. Furthermore ,we also researched the relationship between survivin expression and P53 expression ,and studied the significance of survivin in the carcinogenesis and the development of breast carcinoma. method:It is used to examine the expression of survivin and P53 protein in 7 cases of normal breast tissue ,10 cases of simple hyperplasia (stage â…  ), 20 cases atypical hyperplasia (including 10 cases of stage â…¡ and 10 cases of stage â…¢) and 36 cases of breast cancer tissue (including 22 cases of invasive ductal cancer ,10 cases of adenocarcinoma ,3 cases of medullary carcinoma and 1 case of invasive lobular carcinoma) by using the immunohistochemical Avidi-Biotin complex techniqe.Further more,it was investigated for the correlations with the feature of clinical pathology and the relationships between survivin expression and P53 protein expression. Result:1. The positive expression rate of survivin gene in normal breast tissue , simple hyperplasia,laryngeal atypical hyperplasia and breast carcinoma were 0.00%,10.00%,65.00%,83.33%. there was no difference between the normal breast tissue group and the simple hyperplasia group (P>0.05).The difference was significant when normal breast tissue group was compared with laryngeal atypical hyperplasia group and breast carcinoma group ( P<0.05).The positive rate of survivin in breast carcinoma was significantly higher than those in other groups(P<0.01),and the positive rate of survivin in laryngeal atypical hyperplasia group was significant than that in normal and simple hyperplasia groups(P=0.01).2. The positive expression rate of P53 gene in normal breast tissue , simple hyperplasia.laryngeal atypical hyperplasia and breast carcinoma were:0.00%,0.00%,10.00%,47.22%. The positive rate of P53 in breast carcinoma was significantly higher than that in normal and simple hyperplasia groups ( P<0.001), it was also significant when comparing with laryngeal atypical hyperplasia group ( P=0.01). there was no difference among the normal breast tissue group -< simple hyperplasia group and laryngeal atypical hyperplasia group (P>0.05)o3. In 36 cases of breast cancer, The positive expression rate of survivin gene in clinical stage MI was 81.82%,and the rate that in stage III-IV was 85.33%,the clinical stage III-IV was higher than that in stage I-II,but there was no significant difference between them (P>0.05). The positive expression rate of P53 gene in stage I-II was 36.36%,and the rate that in stage DI-IV was 64.29%,it was same to the positive rates of survivin and P53 protein (P>0.05).4. In 36 cases of breast cancer, The positive expression rate of survivin gene in invasive ductal cancer tissue was 86.36%,and other type of breast cancer were 78.35%, there was no significant difference between them (P>0.05). The positive expression rate of P53 gene ,there was no significant difference between invasive ductal cancer group and other type of breast cancer group too(P>0.05).5. In 36 cases of breast cancer, The positive expression rate of survivin gene in the group with lymph node metastasis and the group without lymph node metastasis were 90.91% and 71.43% respectively, there was no significant difference between them (P>0.05). The positive expression rate of P53 gene in the group with lymph node metastasis and the group without lymph node metastasis were 54.55% and 35.71% respectively,but there was also no significant difference between them (P>0.05).6.1n breast cancer.the function of survivin gene was correlated with P53 gene and the coincidence rate of the positive expressions of them was 56.67%(17/30) ( P<0.05).The survivin expressions were found to be positive in all P53-positive cases,the P53 is found to be negative in 13 survivin-positive cases.
