Objective: To study the antipyretic effect and the mechanism of yin- huang micro-enema(YHCE),we used dry yeast to make fever model and investigated the expression of COX-1 and COX-2 in the hypothalamus tissues of febrile rats after coloclysising with YHCE by real time fluorescence quantitative polymerase chain reaction and immunohisto- chemical methods.This will provid experimental evidence and theoretic base for its clinical use. Methods: 1.The experimental research on antipyretic effet of YHCE: Healthy male Wistar rats,weighting 190±10g.All rats were kept in the experimental environment for 3 days before the experiment. After measuring the basal body temperature of all rats for 3 days,we selected the qualified rats and devided them into several groups randomly.We took the mean value of two temperature before experiment as the basal body temperature.Except the control group,all rats were injected 20% suspension of yeast hypodermically 10m1/kg.One hour after injecting,the coloclysising drugs were administered respectively.Then we observed the body temperature change of the all groups and noted every 1h until 7h. 2.The experimental research on antipyretic mechanism of YHCE: The method was the same as the first part.The rats were decapitated quickly and were taken out hypothalamus tissues every two hours after administered.Then the hypothalamus tissues were stored for the experiments of the real-time fluorescence quantitative polymerase chain reaction and immunohistochemistry.The two experiments were performed according to their kit description. Results:1.The experimental research on antipyretic effet of YHCE: Compared with the basal body temperature,the body temperature of the control group had no obvious change in the two experiments(all P>0.05).After injected 20% suspension of yeast,the body temperature of the distilled water group rised obviously and reached the peak at 6h (all P<0.01).The positive drug group(pediatric paracetamol enema)inhibited markedly the temperature rising from 1h to 5h or 6h after administration(6h P<0.05,else P<0.01).In the experimental research on antipyretic effets of different doses of YHCE,the YHCE high dose group inhibited makedly the temperature rising from 1h to 7h after administration(7h P<0.05,elseP<0.01),compared with the positive drug group,there was no obvious difference between the two groups from 1h to 4h after administration (P>0.05).But the YHCE high dose group was more effective than the positive drug group at 5h after administration(P<0.01).Furthermore,the antipyretic effect of the YHCE high dose group did not disappear until 6h,7h after administration(6hP<0.01,7hP<0.05).The YHCE medium dose group inhibited the temperature rising at 1h, 2h after administration(all P<0.05),but its antipyretic effect was weaker than the positive drug group(P<0.01).Moreover,there was no obvious antipyretic effect of the YHCE medium dose group from 3h to 7h after administration(all P>0.05).The antipyretic effect of the YHCE low dose group had no obvious difference from the distilled water group after administration(all P>0.05).In the experimental research on antipyretic effets of simple recipe and compound recipe of YHCE,the antipyretic effect of the YHCE group was similar to the first experiment. The baical skullcap root group inhibited markedly the temperature rising from 1h to 5h after administration(5h P<0.05,elseP<0.01),but its antipyretic effect was weaker than the positive drug group(P<0.01).The bupleurum chinense group also inhibited markedly the temperature rising from 1h to 4h after administration(all P<0.01),but its antipyretic effect was weaker than the positive drug group too(all P<0.01).The honeysuckle flower group also inhibited the temperature rising at 3h,5h,7h after administration(3h,5hP<0.01,7h P<0.05),but its antipyretic effect was obvious weaker than the positive drug group at 3h ,5h after administration(P<0.01). 2.The experimental research on antipyretic mechanism of YHCE: Under the normal status, there was a bit expression of COX-1 mRNA and protein in the hypothalamu...
