Baicalin is commonly used in clinical medicine for bringing down a fever,the application has a long history of medicinal demand,with good development prospects.However,the current study of the antipyretic effects of baicalin-based material,mainly using the "separation of analysis" approach,limited to a single screening of the active ingredient, neglecting the theory of traditional Chinese medicine - " the total perspective " and the interactive relations between the active ingredients.The study of Baicalin antipyretic mechanism mainly concentrated in the peak heat of heat in hypothalamus,and less involved in the early heat-induced fever and the matter in peripheral blood during fever. Thus,in the quality control of baicalin,clarify the antipyretic mechanism,etc.yet to be perfected.How to properly use of modern scientific theories and scientific and technological achievements of modern life,used in line with the Chinese medicine theory and characteristics of its own feasibility study of ideas and methods,to clarify the material basis of baicalin and its antipyretic mechanism is a serious problem.1 ObjectiveUnder instruction of the System complexity,using the Grey Reati-onal Analysis to deal with many uncertain factors,combining the experimental results and pharmacodynamics fingerprint to study the baicalin antipyretic efficacy material based material.The hypothesis of "traditional Chinese medicine(Scutellaria baicalensis) systems divide the natural ratio of active ingredients to form the complex may be the role of multi-target effect of the material basis" was advanced.The active ingredients that have more relationship with antipyretic are coming from the effect analysis.The active ingredients with its natural ratio of Baicalin form the matching compound.Then through the antipyretic efficacy of the proposed experiments to verify the hypothesis.To observ the circumference and the main center PGE2,the cox content's change,and the changing in three phase of fever.Discussing what is the basis of the antipyretic material of baicalin and its antipyretic mechanism thoroughly.2 Methods2.1 Theoretical methodsTheoretical research was used to analyze the connotation of TCM and system theory in order to clear the characteristics and the guiding ideology of the material basis of Baicalin antipyretic study,which may propose a feasible way for the study.2.2 Experinental methods2.2.1 Collectibity desingBaicalein and baicalin with its natural ratio of Baicalin form the matching compound.We make the compound as the object of study.Then through the antipyretic efficacy of the proposed experiments to compare their results with the Hebei raises mellowly,to verify whether it can represent from different origin,different extracts of the antipyretic effect of screening out good Baicalin Hebei raises mellowly,and that the antipyretic Baicalin material foundation.And then observing the content's changing of PGE2 and cox in circumference and the main center,also the changing in three phase of fever.Discussing what is the antipyretic mechanism thoroughly.2.2.2 Baicalein and baicalin in the formation of the natural ratio of mathing complex antipyretic efficacy of the experimental2.2.2.1 Experiment animalWistar male rats,weighted 190-210 gram.The rats which body temperature were qualify were randomly divided into groups as Baicalin,Baicalein,complex,Hebei Baicalin ethanol extract,aspirin,model,normal control.Do a formal experiment after adapting the operation of rectal temperature measurement.2.2.2.2 Test methodsObserving the body temperature of rats in each group after injecting the dry yeast solution. Planning the average temperature curve.Calculating the maximum high temperature(△T) and the thermal response index(TRI11),to reflect the intensity of fever and duration of effect.2.2.2.3 Analysis methodsUsing SPSS for statistical analysis,such as variance test.2.2.3 Baicalein and baicalin in the natural ratio of mathing complex formation mechanism of the antipyretic effect2.2.3.1 Experiment animalWistar male rats,weighted 190-210 gram.The rats which body temperature qualified were randomly divided into groups as complex,aspirin,model,normal control.Each group was divided into 3h,9h two subgroups.Do a formal experiment after adapting the operation of rectal temperature measurement.Draws blood,the hypothalamus in 3h,9h.2.2.3.2 Test methodsObserving the body temperature of rats in each group after injecting dry yeast solution. Calculating the maximum high temperature(△T).Early fever(model after 3h) and the peak heat(after the model 9h) in peripheral blood and hypothalamus in the determination of PGE2 content using radioimmunoassay;fever peak(after the model 9h) in the hypothalamus determination of cox-1,cox-2 using RT-PCR.2.2.3.3 Analysis methodsUsing SPSS for statistical analysis,such as variance test.3 Results3.1 Results of theoretical studyThe study concluded that baicalin is a complex system.The characteristics of the study of the antipyretic effect of baicalin material basis are systemic,complex.So revealing the material basis for its antipyretic shoud based on the overall concept of TCM and System Theory.So we accout that the antipyretic effects of baicalin material is a internal system of the baicalin systems.Then we proposing that the ratio of the original material,active ingredients differentiated from baicalin,may be the material basis for its antipyretic effect.3.1 The characteristics of the body temperature of rats after injecting dry yeast solutionRat fever changes occur in three phases - rising phase,fever peak,declining phase,which is consistent with the clinical features of fever.Before the three-phase,dry yeast-induced fever in rats there is a drop in body temperature stage,which is characteristic of dry yeast with fever caused.3.2 In the course of fever,baicalin,baicalein,natural ratio of mathing complex formation, Hebei Baicalin ethanol extract showed a significant antipyretic effect,and have different characteristics:Baicalin has the antipyretic effect in the rising phase of fever;Baicalein in the declining phase;the natural ratio of mathing complex formation,Hebei Baicalin ethanol extract in the three phases,and the compound has the similar ability of antipyretic effect and inhibit the heating strength in fever peak with Hebei Baicalin ethanol extract.So natural ratio of the complexe has the similar characteristics and the role of fever with the Hebei Baicalin ethanol extract,to a certain extent Baicalin can represent the material basis of antipyretic effect.In the certain extent it may be the antipyretic material basis of baicalin.3.3 There are some features of peripheral blood and hypothalamus of PGE2,cox-1,cox-2 content of dry heat-induced fever in rats in the three-phase.In the early period of heat,the content of PGE2 in peripheral blood is high,and in the hypothalamus is low;in the fever peak,the content of PGE2 in peripheral blood is low,and in the hypothalamus is high;and in the peak heat,cox-1,cox-2 levels in the hypothalamus were significantly increased.It showing that in the fever peak,the high content of PGE2 in peripheral blood carriered with protein binding with the blood through the blood-brain barrier to reach the hypothalamus caused by increased body temperature.In the peak heat,the content of cox-1,cox-2 increased and synthesis of PGE2 more to up the temperature point.3.4 Baicalin and baicalein natural ratio of the mathing complex formation showed significant ability of inhibit the content of PGE2 both in peripheral blood and hypothalamus in fever peak,but there is no significant inhabit ability in the early heat period;At the same time,the compound also has ability of inhabiting the levels of cox-1 and cox-2 in the hypothalamus in fever peak,with a considerable trend of positive control drug aspirin.In addition,we found that,different with the natural ratio of the mathing complex formation, the content of PGE2 in peripheral blood of aspirin control showed no inhibitory effect in fever peak.We speculate that baicalin and baicalein natural ratio of the mathing compound inhibited the increasing of PGE2 content both in the peripheral blood and hypothalamus.Its antipyretic effect may be to inhibit the peak heat of the heat intensity,and related with peripheral blood and hypothalamus of PGE2.The compound brings down a fever in peak by reducing cox-1 and cox-2 content m the hypothalamus,inhibitting the increase of PGE2. So the compound showed a significant lowing body increased temperature in the peak period.4 Conclusions4.1 Chinese medicine is a highly complex system of chemical substances.Baicalin is a complex system.The material basis for efficacy of baicalin is a intersystem of baicalin system.The research of material basis for efficacy of traditional Chinese medicine should under the knowledge of the system complexity and systems thinking,doing the comprehensive and systematic study.4.2 Experimental results show that the working hypothesis - "traditional Chinese medicine (Scutellaria baicalensis) systems divide the natural ratio of active ingredients to form the matching complex may be the role of multi-target effect of the material basis" has a certain scientific.4.3 In accordance with the preparatory work for different places,different extraction methods of the antipyretic effect of screening out good Hebei ethanol extract of Scutellaria, the antipyretic effect associated with higher levels of baicalin,baicalein natural ratio of the matching complexs formed with Hebei ethanol extract Baicalin similar characteristics and the role of fever,to a certain extent Baicalin can represent the material basis of antipyretic effect.4.4 Baicalin and baicalein natural ratio of the matching complex form may reduce the levels of PGE2 of hypothalamus and peripheral blood to antipyret fever.The Antipyretic Role of the natural ratio——mathing complexs may relate with PGE2,cox-1 and cox-2 in peak fever.
