Objective 1.Explore the biological features of adult rat bone marrow-derived mesenchymal stem cells(MSCs). 2. MSCs were induced into neural-like cells . 3.Produced Parkinson's disease rat. 4. We transplanted adult rat MSCs into the striatum of Parkinson's disease (PD) rat and examined their survival, differentiation, and the behavior changes in PD rats, which will provide a theoretical foundation and technical method for clinic PD therapy by stem cells.Methods 1. Pured adult rat bone marrow-derived MSCs be plated into culture plate. The growth characteristic of MSCs was measured in passage cultures. Pured MSCs were induced into neural-like cells by incubation with basic fibroblast growth factor ( bFGF ) and N2 in DMEM/F12 medium. Immunocytochemistry staining were performed. Cell cycle was analyzed in MSCs and MSCs-dirved cells by flow cytometry(FCM). Using patch- clamp whole - cell recording to search for electrophysiological characteristics of MSCs and MSCs-dirved cells. 2. Produced Parkinson's disease rat by direct unilateral injection of 6-OHDA into the rat brain medial forebrain bundle(MFB). 3. 15 PD rats were selected and divided into two groups,a Brdu-labeling MSCs transplant (groupn= 11) ,a control phosphate-buffered saline (PBS) transplant group (n= 4). Transplantions were stereotaxically transplanted into the striatum of Parkinson disease (PD) rat. Apomorphine induced rotation was monitored each 2 weeks after transplantation.The transplanted cell positive for tyrosine hydroxyase(TH) and Brdu was analyzed in 2nd and 16th week respectively.
