Objective: To study the effect of "Yiqitiaoxue, fubenpeiyuan" Acup Therapy on selective neuron lost of Senile Accelerated Mouse (SAM) P8, a model of AD, treated with acupuncture. Observe the number and sharp of neuron in the hippocampus and the cortex of temporal lobe, activation and accrementition of horizontal cell, as well as the expression of apoptosis relative protein. Discusse the possible mechanism of acupuncture in neuroprotective effect.Methods: Altogerher 48 mice of seven-month old male SAMP8 were selected and allocated randomly to three groups: P8 control group, sham-acupuncture group and acupuncture group. Moreover, 16 age- and sex-matched SAMR1 mice were enrolled as homology control group. The acupuncture group was given the treatment of "Yiqitiaoxue, fubenpeiyuan" Acup Therapy once daily for 14 days. At the end of the study, ten mice in each group were sacrificed and their brains were dissected, weighed, measured the length and the width. The other mice were transcardially perfused with 0.9% NaCl followed by 4% paraformaldehyde. Paraffin sections were stained by toluidine blue technique to observe tigroid body and the neuron number, the neuron sharp, research the expression of GFAP, Bax, Bcl-2 in the CA1 and CA3 field of the hippocampus and cortex of temporal lobe by histoimmunechemical. Semiquantitative analysis is done with image analysis system and all data analyses were performed with SPSS11.0. Results:1. The whole brain's length, width and weight of SAMR1 exceeded that of SAMP8 control group, but without significant differences were found between the two groups. The neuron shape of the cerebrum for P8 control group occurred degenerative alteration.
