OBJECTIVE To investigate the influence of acupuncture treatment on the expression of high mobility group protein box 1 (HMGB1) and glial fibrillary acidic protein (GFAP) in hippocampus CA1 area of Senescence Accelerated Mouse SAMP8, And to explore the possible mechanism of treating AD by acupuncture.METHODS 20 five-month old SAMP8 mice were randomly divided into behavioral testing acupuncture treatment group and model group, and select 5 months of normal aging mice (SAMR1) 10 only as a normal control group.Acupuncture treatment group received Baihui, Shenshu, xuehai and geshu, the model group and control group on the same day from the date of capture and fixation.4 weeks later, the hippocampus of mice were taken out to examine the expression of HMGB1 and GFAP by immunohistochemistry method.RESULTS 1.Behavioral results:The 5 months SAMP8 mice had significantly decreasad in ability of learning and memory than SAMR1 mice; 2.The results of immunohistochemistry:(1)compared with the normol control group,the IOD of HMGB1 in model group was increased (P<0.05);the IOD of HMGB1 in Acupuncture treatment group campared with the modol group was decreased(P<0.05);(2)compared with the normol control group,the IOD of GFAP in model group was increased (P<0.05);the IOD of GFAP in Acupuncture treatment group campared with the modol group was decreased(P<0.05).CONCLUSION 1.SAMP8 is reliable as a animal model of Alzhiheimer's.2.The level of HMGB1 and GFAP in the SAMP8 mice were increased, which probably is part of pathology of Alzheimer's disease.3.The acupuncture treatment in bushen huoxue methods can improve the learning and memory ability in SAMP8, and by inhibiting the grow of glial cells, suppression of the immuneinflammatory reaction on the nerve cell injury, to achieve the treatment of AD。...
