Objective: The study aims to observe the effect of apelin-13 to the vasoconstriction and relaxation of spontaneously hypertensive rat`s aorta. To approach the possible signaling pathway of NO and ERK1/2, which could mediate the apelin-13`s vasoconstriction and relaxation reaction of spontaneously hypertensive rat`s aorta. Methods: Take out of thoracic aorta after execute the spontaneously hypertensive rats and Wistar-Kyoto rats. Use the means of vaso-culture and Power-Lab system to determination the change of tension on rat`s vascular rings, which could be influenced by apelin-13, L-NNA and PD98059.Results: Exogenous apelin-13 have the action of blood vessel endothelium`s depend relaxation. The signaling pathways of nitric oxide and erk1/2 are all acts as real roles in apelin-13.Conclusion:In our study, we found that exogenous apelin-13 can relaxed the blood vessel. Accompany with the hypertension, the obstacle of nitric oxide signaling pathway of apelin-13 induced the effect weaken of apelin-13 which can relax blood vessel. At the same time, the effect of blood vessel relaxation of apelin-13 is related with erk1/2 signaling pathway.
