objective: To investigate the effects of lipoxygenase inhibitorson the proliferation and apoptosis of HepG2 cells and its relevant mechanism.Methods: human hepatoma cells line HepG2 trained outside the body,the effects of lipoxygenase inhibitors (nordihydroguaiaretic acid; NDGA ) on the growth and survival of human hepatoma cells line HepG2 were determined with microculture tetrazolium assay (MTT), growth curve and colony-forming assay and apoptosis and cells cycle analysis was determined using HE staining and flow cytometry. Measured the protein expression of Caspase~3, PCNA with immunohistochemistry. Western blot analysis were employed to observe the effec to NDGA on the expression of wtp53 and c-erbB-2 protein.Results: NDGA had marked suppressive effect on the growth of HepG2cells line.The cells were treated with varies concentrations of NDGA(50, 100, 150, 200)μmol/l for up to 72 hr Cells proliferation was measured using MTT. The inhibitory rates of the cells were (27.34± 5. 95)%, (41. 76 + 7. 47)%, (57. 08 + 4. 36)% and (69. 17 + 4. 42)% respectively in 72 hours. NDGA had a remarkalbe cytotoxic effect to HepG2 with IC50 of almost 109. 13μmol/L. Colony-formating rate of HepG2 cells were (21.63 + 0.70)%, (17. 33 + 0. 96)%, (12.53 + 0.45)%, (7. 97 + 0. 92)% respectively, which were much lower than that of control group (31. 70 + 0. 62)%, (P<0. 01). At the same time, Flowcytometry revealed NDGA could induce the apoptosis of HepG2 with the cells cycle arrested on G, phase.
