Objective : The aim of the present study was to determine effects of hepatocyte growth factor (HGF) on the gowth and apoptosis of human liver cancer cell line HepG2, and mechanism behind them. This study also means to investigate if there is synergistic effects of HGF plus pirarubicin (or theprubicin, THP) on the proliferation and apoptosis of HepG2 cells, to explore the value of HGF in the therapy against hepatocellular carcinoma and to offer the feasibility and corresponding theory evidences for subsequent animal and clinical experiment. Methods: The effects of HGF on the growth of HepG2 cells was evaluated by MTT assay; the expression of proliferating cell nuclear antigen (PCNA) was assessed by immunocytochemical method; apotosis index(AI) was counted by fluorescent staining of the hepatocye nuclei with Hoechst33258; the apoptotic rate (AR) was quantified by flow cytometry (FCM). The flow cytometry (FCM) also was used to detect phase distribution of the cycles and RT -PCR technology for the epression of p16 gene. Results: 1: The data of MTT showed that HGF significantly suppressed the growth of HepG2 cells in a time and dose-dependent manner.
