In chiral recognition process polysaccharide-derivatized as a chiral selector, exhibits high selectivity , high efficiency and wide range of application. This paper reports the separation of the enantiomers of ten pairs of compounds on a derivatized amylose stationary phase by normal-phaseliquid chromatography .we learn the mechanism about the chiralpakAD column by above results.This paper reports the separation of the enantiomers of ten pairs of compounds on chiralpakAD column by normal-phaseliquid chromatography .8 racements achieved enantiomeric resolution at different level. The result shows that the molecules with C=O, -N-, -S-, -O- can get considerable enantioselectivity on this CSP. chiralpakAD possess high-order configuration ,so it have chiral cavum. We also discuss effects of column temperature ,the alcoholic modifier , additive and velocity of flow on the separation of the enantiomers.Proceed from physical chemistry angle, study the chiralpakAD column in terms of thermodynamics and dynamics. Proceed from foundation of physical chemistry and find the van't Hoff plot for the enantioselectivity factor for data taken when heating the column from 5 to 50°C was not superimposable with the same plot prepared with data from the cooling process 50 to 5°C.This observation suggested that the stationary phase was undergoing a thermally induced irreversible conformational change that altered the separation mechanism between the heating and cooling cycles.
