【Objective】To investigate the effect of Emodin on inducing humanacute myeloid leukemia cell line HL-60 apoptosis and proliferationinhibition and on expression of related protein and mRNA .【Methods】HL-60 cell was exposed to verious dosages of Emodin.Proliferation inhibition was detected by MTT assay and clone formation assay ,cell apoptosis was observed by flow cytometry,TUNEL labeling method ,DNAfragmentation electrophoresis and mitocapture apoptosis detection. Theexpression of c-myc ,pim-2,bcl-2,mcl-1 and bax gene was detected by RT-PCRand C-myc and Bcl-2 protein was detected by Western-Blot.【Results】The results showed that Emodin remarkably inhibited thecell proliferation .The IC50 value was 20uM. Apoptosis in HL-60 cells couldbe efficiently induced by Emodin in a dose dependent manner and cells werearrested at G0/G1. The results of RT-PCR and or Western-Blot showed c-myc,pim-2 was decreased after Emodin treatment, while bcl-2,mcl-1,bax remainunchanged.【Conclusion】Emodin could efficiently induce growth inhibition andapoptosis in HL-60 cells .C-myc and pim-2 may be involved in this process.Emodin has no effects on the expression of bcl-2,mcl-1 and bax.
