Objective: On the basis of previous study of the apoptosis pathways, To investigate the effects of emodin on expressions of apoptosis inducing factor (AIF) and endonuclease G (EndoG) mRNA in rat CBRH-7919 cells and its mechanism in inducing apoptosis. After that to investigate the anti-tumor effect and mechanism of emodin.Methods: (1) The rat CBRH-7919 cells were cultured in the medium.(2) The rat CBRH-7919 cells were treated by emodin in various concentrations and different time points respectively; and then the cell proliferation was assayed by MTT method, meanwhile, the IC50 were computed at every time point. (3) The CBRH-7919 cells were divided into 4 groups: blank control group, Caspase inhibitor Z-VAD-FMK group, Emodin group, Emodin and Z-VAD-FMK group. (4) TUNEL were used to detect the apoptosis.(5) The expression of AIF and EndoG mRNA were examined by Real time PCR.Results: Compared with the control group, emodin obviously increased the inhibitory rate of CBRH-7919, and had dose-dependent effect and time- dependent effect. And the IC50 (48h) of the CBRH-7919 were 44.8μmol/L. After the treated by emodin or emodin and Z-VAD-FMK, it showed a typical apoptotic morphology. The expression of AIF and EndoG mRNA significantly increased in emodin or emodin and Z-VAD-FMK groups than that in the blank group(P < 0.01).Conclusion: Emodin could induce the apoptosis of CBRH-7919 lines in vitro by up-regulating the expression of AIF and EndoG , which can induce apoptosis through the caspase-independent pathway.
