Experimental Study On Apoptosis Of Human Lung Cancer Cell Line SPC-A-1 Induced By Solanum Lyratum Thunb Extract

Objective:to study the effect of Solanum Lyratum Thunb extract on human lung cancer cell line and its possible mechanism. Method: MTT detect the effect of growth of Solanum Lyratum Thunb extract on human lung cancer cell line;inversion microscope and fluorescenece staining microscope were used to detect the morphological changes of SPC-A-1 cells;to detect DNA ladder Agarose gel electrophoreris was applied;to detect the cell apoptosis rate and to analyse the cell cycle of SPC-A-1 and to detect the expression of bcl-2 gene in SPC-A-1 cells FCM was performed. Results: Solanum Lyratum Thunb extract could inhibit SPC-A-1 cell growth,its effect is related with the concentration and action time of Solanum Lyratum Thunb extract;SPC-A-1 cells treated with Solanum Lyratum Thunb extract for 48h showed early changes of apoptosis;Agarose gel electrophoreris took on typical DNA ladder;SPC-A-1 cells showed apoptosis mountain and apoptosis rate depended on concentration,DNA synthesis was reduced ,bcl-2 expression was decreased. Conclusion: a certain concentration of Solanum Lyratum Thunb extract can inhibit the growth of SPC-A-1 cells and can induce apoptosis of SPC-A-1 cells.Its action mechanism is related with bcl-2 gene.
