ObjectiveTo explore the expression of nestin and VEGF and the effect of exogenous bFGF on nestin and VEGF expression in hypoxia- ischemia brain of neonatal SD rat .Investigate pretection mechanism of bFGF on hypoxia- ischemia brain of neonatal SD rat. The result could be used as evidence to understand generally of the pathogenesis of HIBD and the reason for our clinic therapy. MethodsWe sorted eighety-four neonatal SD rats in three groups , 28 in each group, respectively a shamoperated group,in which the rats were give false operation;a bFGF group, bFGF 4ug/Kg i.p for 7days following hypoxic-ischemia and a HIBD group,vehicle were given i.p for 7days. The HIBD models were established with 7 day old SD rats. We used the immunohistochemical ABC method to detect the expression of nestin and VEGF at 3h, 12h, 24h, 48h, 72h, 168h, 336hours in the brain after ischemia and hypoxia and we deal the data statistically with One-Way ANVOA . Results(1) Nestin was expressed weekly in the hippocampus and extraventricular zone and not expressed in the cortex of the shamoperatedgroup. VEGF was not expressed in the rat brain of the shamoperated group.(2) it is obviously that the number of nestin and VEGF positive cells increased in the rat brain after ischemia and hypoxia compared with the shamoperated group and varied with time. The time peaks of the nestin expression are at 7days after ischemia and hypoxia. The time peaks of the VEGF expression are at 12hour after ischemia and hypoxia.(3) Exgeonous bFGF enhanced the expression of nestin and VEGF in the hypoxia- ischemia brain of the neonatal rat .The nestin signals in bFGF groups are significantly higher than HI groups at lday(p<0.05) and persistent to 14days. .The VEGF signals in bFGF groups are significantly higher than HI groups at 3hour (p<0.05) and persistent to 3days. ConclusionThe number of nestin and VEGF positive cells increased in the rat brain after ischemia and hypoxia conpared with the shamoperated group. Exgeonous bFGF could enhanced the expression of nestin and VEGF in the hypoxia- ischemia brain of the neonatal rat .Indecated that the bFGF may play important roles in restoration neurons damaged by hypoxia- ischemia.
