Effect Of Ethanol Extract Of Three Kinds Of Bulb Fritillariae Cirrhosae On Guinea Pigs With Allergic Asthma

Bulbus fntillariae cirrhoosae is one of the rare Chinese traditional medicine. It's bulb has many comprehensive pharmacologic effects.Experiments were carried on to research the effects of FW (F. wabuensis S. Y. Tang et S. C. Yueh), FM (F. mllea S. Y. Tang et S . C. Yueh) cultivated and FU (Fritillaria unibracteata Hsiao et K. C. Hsia var. longinectaria S. Y. Tang et S. C. Yueh) on Pulmonary Mechanical Function in Sensitized Guinea Pigs. The effects of drug on airway were evaluated by determing the changes of pulmonary Resistance (RL) and pulmonary dynamiccompliance (Cdyn) by pneu-motachography. Salbutamol Salbutamol Sulfate is used as contrast; To investigate the effects of ethanol extract of FW and FM cultivate -d on the strips of ileum isolated from sensitized Guinea Pig by the Schultz-Dale experiment. Dexamethasone is used as positive contrast.Results:1. Ethanol extract of FW(20mg/kg. 40mg/kg)inhibt the reduction of Cdyn of ovalbumin-sensitized Guinea Pigs induced by antigen attack. P<0.05 compared with model group at 1,3,6 and 12 minute after antigen attack. Ethanol extract of FM also can inhibt the reduction of Cdyn of ovalbumin-sensitized Guinea Pigs induced by antigen attack.FM(10mg/kg) P<0.05 compared with model group at 1 and 12 minute after antigen attack. FM(40mg/kg) P<0.01 compared with model group at 1 and 6 minute after antigen attack. P<0.05 compared with model group at 0,3,9 and 12 minute after antigen attack.There is no significant changes of FU on Cdyn .2. Three kinds of Ethanol extract can significantly inhibit the increase of RL of ovalbumin-sensitized G uinea P igs induced by antigen attack. P < 0.01 compared with model group at 1 minute after antigen attack. FM and FW(20mg/kg^ 40mg/kg) have better effect. P>0.05 compared with Salbutamol Sulfate group. The difference between FW and FM (middle dosage and high dosage) has no statistical meaning.3. Three kinds of Ethanol extract have no significant effects on the increase after antigen attack and the reduction of breath frequency of sensized guinea pigs.4. Ethanol extract of FW and FM (6.67x10°mg/ml and 2.67><10"3mg/ml) can inhibit the contractions of ileum of guinea pigs in vitro significantly induced by antigen attackyP< 0.01 compared with model group. Inhibition effects of FM (low dosage group) is weaker than other groups. P<0.0l compared with other groups. The difference between Dexamethasone andthe two kinds of ethanol extract has no statistical meaning.Conclusion:1. Ethanol extract of FW and FM cultivated possess inhibit effects on the deduction o f C dyn a nd t he i ncrease o f R L of s ensitized G uinea P igs induced by antigen attack.2. Ethanol extract of FW and FM cultivated possess inhibit effects on the contractions of ileum of guinea pigs induced by antigen attack in vitro.3. FW and FM can alleviate the allergic asthma symptoms induced by antigen attack repeatly.
