Objective:To study on the treatment of acute bronchitis of Bulbus Fritillariae Cirrhosae cough relieving granule and reveal the mechanism of Bulbus Fritillariae Cirrhosae cough relieving granule in treatment of acute bronchitis.Methods:Variety of experimental techniques methods used in the experiments, from whole organs, multi-level, molecular level, a comprehensive system of in-depth evaluation of role characteristics and mechanism in the treatment of acute bronchitis of Bulbus Fritillariae Cirrhosae cough relieving granule.To build the animal model of acute bronchitis with the method of cigarettes and sawdust and study the treatment of acute bronchitis of Bulbus Fritillariae Cirrhosae cough relieving granule. After the experiments, observing the ultrastructure changes of bronchus of rats and mice with Transmission electron microscopic, using Elisa method for determination of NF-kB, sICAM-1, IL-6, IL-8, TNF-αin serum of rats and IL-8, TNF-αin serum of mice. From the perspective of histopathology, ultrastructural, and molecular to evaluate of effect of traditional Chinese medicine compound formula in treatment of acute bronchitis.To investigate the cough relieving effect of Bulbus Fritillariae Cirrhosae with methods of ammonia water inducing cough in mice and citral inducing cough in guinea pig and capsaicin inducing cough in guinea pig; using Elisa method for determination of SP in serum of guinea pig and SP in bronchoalveolar lavage fluid of guinea pig. To investigate the antiasthmatic effect of Bulbus Fritillariae Cirrhosae with methods of acetylcholine/histamine mixture inducing asthma and the method of isolated guinea pig trachea spiral strip.To investigate the phlegm expelling effect of Bulbus Fritillariae Cirrhosae with methods of phenol red phlegm expelling in mice and the method of glass tube phlegm expelling in rats, using HPLC method to determine the DPPC level of bronchoalveolar lavage fluid. To investigate the antibacterial effect of food with the methods of in vivo/in vitro and the Anti-inflammatory effect of food with the methods of xylene inducing auricular swelling in mice, fresh egg white inducing voix pedis swelling in rats, cotton ball granuloma in miceResults:1,The study of the treatment on rats acute bronchitis of Bulbus Fritillariae Cirrhosae cough relieving granule has shown:Compared with model group, less or small amount of inflammatory cell infiltration of rats bronchus could be seen under light microscope and the IOD and target area of inflammatory cell infiltration significantly decreased in the groups of high dosage, middle dosage and low dosage; mitochondrial of bronchus ciliated epithelium shows normal status in the groups of high dosage and middle dosage but mild swelling in the low dosage group;the level of IL-6,IL-8,TNF-α,NF-kB,sICAM-1 could be significantly decreased in the groups of high dosage and middle dosage, the level of TNF-α,NF-kB,sICAM-1 could be significantly decreased in the groups of low dosage.2,The study of the treatment on mice acute bronchitis of Bulbus Fritillariae Cirrhosae cough relieving granule has shown:Compared with model group, less or small amount of inflammatory cell infiltration of rats bronchus could be seen under light microscope and the IOD and target area of inflammatory cell infiltration significantly decreased in the groups of high dosage, middle dosage and low dosage; the level of IL-8 could be significantly decreased in the groups of high dosage and middle dosage and low dosage, the level of TNF-αcould be significantly decreased in the groups of high dosage and middle dosage.3,The study of the cough relieving effect of Bulbus Fritillariae Cirrhosae cough relieving granule has shown:Compared with blank group, the latency of cough of mice inducing by ammonia water could be significantly increased in the groups of middle dosage and low dosage; and number of cough in 2 minutes of mice could be significantly decreased in the groups of high dosage, middle dosage and low dosage.the latency of cough and number of cough in 5 minutes of guinea pig inducing by citral could be significantly respectively increased and decreased in the groups of high dosage, middle dosage and low dosage;the number of cough in 5 minutes of guinea pig inducing by capsaicin and the level of SP in guinea pig serum could be significantly decreased in the groups of high dosage, middle dosage and low dosage, and the level of SP in the bronchoalveolar lavage fluid could be significantly decreased in the groups of middle dosage and low dosage.4,The study of the antiasthmatic effect of Bulbus Fritillariae Cirrhosae cough relieving granule has shown:Compared with the blank group, asthma latent period of guinea pig inducing by acetylcholine/histamine mixture could be significantly increased in the groups of high dosage and middle dosage; the contraction tension of the Isolated guinea pig trachea spiral strip inducing by acetylcholine (final concentration:1×10-7g/ml) could be significantly decreased in the groups of high dosage, middle dosage and low dosage.5,The study of the phlegm expelling effect of Bulbus Fritillariae Cirrhosae cough relieving granule has shown:Compared with the blank group, the amount of phenol red of mice significantly increased in the groups of middle dosage and low dosage; the amount of glass tube phlegm expelling of rats significantly increased in the groups of high dosage, middle dosage and low dosage; the level of alveolar surfactant (DPPC) could be increased in the different of groups.6,The study of the antibacterial effect of Bulbus Fritillariae Cirrhosae cough relieving granule has shown:Bulbus Fritillariae Cirrhosae cough relieving granule could play the bacteriostatic action in vitro test to these following gram-negative bacteria and gram-positive bacteria including staphylococcus aureus, kurthia gibsonii,macrococcus;megacoccus, enterococcus faecalis, coryneba cterium, bacillus spp, escherichia coli, enterobacter cloacae, chryseobacterium indologenes, acinetobacter lwoffii, proteus vulgaris, shigella flexneri;Survival rate of mice by intraperitoneal injection of staphylococcus aureus are respectively 33.33%,41.67%,25% in the groups of high dosage, middle dosage, low dosage and survival rate of mice by intraperitoneal injection of escherichia coli are respectively 58.33%,58.33%,41.67% in the groups of high dosage, middle dosage, low dosage.7,The study of the anti-inflammatory effect of Bulbus Fritillariae Cirrhosae cough relieving granule has shown:Compared with the blank group, the swelling of auricle of mice inducing by the xylene significantly reduced and the swelling of voix pedis of rats inducing by the 10% fresh egg white significantly reduced and the weight of cotton ball Granuloma of mice significantly reduced.Conclusion:The characteristics of treatment of acute bronchitis of Bulbus Fritillariae Cirrhosae cough relieving granule are as following:the regulation of mulit-level and multi-steps in inhibition of pathogenic factors and reducing symptoms. The pathological state of in bronchus of mice and rats could be reduced; the level of IL-6, IL-8,TNF-α, NF-kB, sICAM-1 could be decreased;the level of IL-8 and TNF-αcould be decreased. The effect of cough relieving, antiasthmatic, phlegm expelling, antibacterial, anti-inflammatory could be able to play as important role in the treatment of acute bronchitis...
